Single cell mass spectrometry analysis of drug-resistant cancer cells: Metabolomics studies of synergetic effect of combinational treatment

Xingxiu Chen et al.

Summary: The study demonstrates that metformin can enhance the sensitivity of irinotecan-resistant cells and restore the therapeutic effect. Metabolomics analysis reveals that metformin treatment leads to downregulation of lipids and fatty acids, potentially through the inhibition of fatty acid synthase. Combined treatment of metformin and irinotecan shows the highest inhibition of fatty acid synthase compared to mono-treatment.

Purine nucleoside phosphorylase enables dual metabolic checkpoints that prevent T cell immunodeficiency and TLR7-associated autoimmunity

Evan R. Abt et al.

Summary: The deficiency of PNP can lead to both immunodeficiency and autoimmunity. The mechanisms behind this paradoxical association are not fully understood. This study reveals that the inactivation of PNP can affect T cell development and B cell signaling, resulting in immune abnormalities and autoimmune diseases.

Identification of Lipid Heterogeneity and Diversity in the Developing Human Brain

Aparna Bhaduri et al.

Summary: The lipidome, understudied but fundamental to life, was profiled in 154,910 single cells across different developmental ages and brain regions using single-cell mass spectrometry-based protocols, resulting in a unique lipid atlas of the developing human brain. Differentially expressed lipids were identified across brain structures, cortical areas, and developmental ages, enabling the inference of lipid profiles of major cell types and the detection of putative celltype specific lipids from the data set. This data set will facilitate further investigation of the developing human brain lipidome.
