Exosomes Derived Neuronal Markers: Immunoaffinity Isolation and Characterization

Ghada Yousif et al.

Summary: This study optimized an immunoaffinity-based isolation protocol for specific exosomes or neuronally derived exosomes (NDE). The results showed successful separation of CD63+ and L1CAM+ exosomes from serum using functionalized magnetic beads, and isolation of NDE using GluR2/3-capturing antibody. The study suggests that GluR3/2 may be an effective candidate for isolating neuronal exosomes.

CNS endothelial derived extracellular vesicles are biomarkers of active disease in multiple sclerosis

Michael Mazzucco et al.

Summary: This study developed a novel method to identify central nervous system (CNS) endothelial derived extracellular vesicles (EV) in human blood samples, and found that the plasma concentration of CNS endothelial derived EV was significantly increased in patients with active multiple sclerosis (MS), suggesting that it may serve as a biomarker of blood-brain barrier permeability and active disease in MS.

Initiation and progression of α-synuclein pathology in Parkinson's disease

George K. Tofaris

Summary: alpha-Synuclein aggregation is a critical molecular process in Parkinson's disease pathogenesis, and its nature influences the clinical heterogeneity. Alpha-synuclein assemblies need to exhibit seeding competency and toxicity for neurodegeneration to occur. It is currently unknown which alpha-synuclein assemblies are most relevant to the human condition.

Effective Separation of Cancer-Derived Exosomes in Biological Samples for Liquid Biopsy: Classic Strategies and Innovative Development

Yujiao Xie et al.

Summary: Liquid biopsy enables non-invasive detection of cancer-derived components, facilitating clinical diagnosis and surveillance of cancer. The investigation of cancer-derived exosomes, nano-sized vesicles secreted by cancer cells, in liquid biopsy has revealed their important roles in intracellular communication and disease development. This review analyzes classic strategies for exosome separation, discusses their advantages and limitations, presents innovative multi-strategy methods for efficient isolation, and discusses future development trends.

Catalysed amplification of faradaic shotgun tagging in ultrasensitive electrochemical immunoassays

Mohamed Sharafeldin et al.

Summary: We present a novel electrochemical protein quantitation method that involves shotgun biotin tagging of proteins before interfacial immunocapture and polymeric enzyme tagging. The highly amplified faradaic signals produced by a novel ferrocene-tyramine adduct allow for the detection of protein markers at attomolar levels and across a dynamic range of 5 orders of magnitude. This work supports the ultrasensitive detection of protein markers in a single antibody immunoassay format.

Validation of α-Synuclein in L1CAM-Immunocaptured Exosomes as a Biomarker for the Stratification of Parkinsonian Syndromes

Cheng Jiang et al.

Summary: The study demonstrates that measuring alpha-synuclein levels above 14 pg/mL in L1CAM-immunocaptured serum exosomes can robustly differentiate Parkinson's disease from other movement disorders. Furthermore, elevated levels of exosomal clusterin in individuals with 4-repeat tauopathies, combined with alpha-synuclein, improves the assay's performance in distinguishing Parkinson's disease from 4-repeat tauopathies. However, correction for the generic exosomal protein syntenin-1 did not consistently enhance the assay's performance.
