In situ synthesis of Au nanoparticles confined in 2D sheet-like metal organic frameworks to construct competitive PEC biosensor for sensitive analysis of autism spectrum disorder miRNA

Hong-Yan Shan et al.

Summary: Autism spectrum disorders (ASDs) are a significant concern worldwide, and miRNA detection using the photoelectrochemical (PEC) method has attracted attention. In this research, 1,2,4,5-benzenetetramine tetrahydrochloride (BTA) was used as a ligand to form AuCl4- -MOFs, followed by the formation of Au NPs embedded in MOFs (MOFs@Au NPs). The MOFs@Au NPs showed significantly increased anode photocurrent compared to Au NPs alone. A label-free PEC biosensor was constructed using MOFs@Au NPs as a photosensitizer to detect miRNA-23a-3p related to ASDs. The sensor exhibited a wide linear range and low limit of detection, making it promising for blood-based RNA detection in biomedicine.

One-step isothermal amplification strategy for microRNA specific and ultrasensitive detection based on nicking-assisted entropy-driven DNA circuit triggered exponential amplification reaction

Changjing Yuan et al.

Summary: Early diagnosis of cancers such as pancreatic cancer, which has atypical symptoms and high mortality, is crucial. Exponential amplification reaction (EXPAR) is an effective isothermal amplification method for detecting microRNAs (miRNAs), but it lacks specificity. To address this issue, a nicking-assisted entropy-driven DNA circuit triggered exponential amplification reaction (NAED-EXPAR) is proposed.

An RNA-based catalytic hairpin assembly circuit coupled with CRISPR-Cas12a for one-step detection of microRNAs

Pinru Chen et al.

Summary: In this study, a one-step detection method for microRNAs based on the RNA-based catalytic hairpin assembly (CHA) circuit coupled with CRISPR-Cas12a was proposed. The target miRNA can specifically trigger RNA-based CHA and induce a configurational change of the blocked crRNAs into precursor crRNAs (pre-crRNAs), which can be processed into mature crRNAs to function by leveraging the inherent RNase activities of Cas12a. The developed circuit achieves a femtomolar detection limit and shows accurate detection of miRNA levels in different cell lines.

Genetically Encoded Sensor Enables Endogenous RNA Imaging with Conformation-Switching Induced Fluorogenic Proteins

Wen-Jing Zhou et al.

Summary: Genetically encoded molecular tools are crucial for live cell RNA imaging, and this study develops a new sensor system using conformation switching RNA induced fluorogenic proteins that enable multicolor imaging and signal amplification of endogenous RNAs. The sensor is designed with an RNA sensing module and a degron-fused fluorescent protein reporter for high-contrast imaging of RNA abundance and dynamics in live cells. Furthermore, the sensor system is extended to a multicolor palette and engineered into a signal amplifier, providing a valuable platform for RNA imaging in biomedical research and clinical theranostics.
