Clinical and Histopathological Features of Gelsolin Amyloidosis Associated with a Novel GSN Variant p.Glu580Lys

Maja Potrc et al.

Summary: Gelsolin amyloidosis is a rare genetic disease caused by mutations in the GSN gene, commonly presenting with corneal lattice dystrophy, loose skin, and heart arrhythmia. A novel GSN variant p.Glu580Lys was associated with optic nerve impairment in addition to typical phenotypes. Histopathological analysis showed amyloid deposits in the skin, suggesting the mutation affects protein folding in the fifth domain of gelsolin protein.

A novel GSN variant outside the G2 calcium-binding domain associated with Amyloidosis of the Finnish type

Sean Mullany et al.

Summary: This study reports a novel association between the predicted pathogenic GSN:c.1477T>C variant and amyloidosis of the Finnish type, and is the first to provide functional evidence of a pathological GSN variant at a locus distant to the critical G2 calcium-binding region, resulting in the phenotype of amyloidosis of the Finnish type.
