A single-cell transcriptomic atlas tracking the neural basis of division of labour in an ant superorganism

Qiye Li et al.

Summary: Using single-cell transcriptomics, this study characterized the brain cell repertoire of the pharaoh ant and identified changes in cell composition that underlie division of labour and reproductive specialization. The study found that male and worker ant brains have opposite trends in cell composition, while the composition of gyne and queen brains remained generalized. Role differentiation from virgin gynes to inseminated queens induced abundance changes in roughly 35% of cell types.

Single-cell transcriptomic analysis of honeybee brains identifies vitellogenin as caste differentiation-related factor

Wenxin Zhang et al.

Summary: The study found that honeybee queens and workers have similar genomes but occupy different caste statuses. By establishing single-cell transcriptomic atlases of honeybee brains, five major cell groups were identified, and it was observed that vitellogenin is highly expressed in specific cell types in queen bees.

Redefining Microglial Identity in Health and Disease at Single-Cell Resolution

Francesca Provenzano et al.

Summary: Recent development in single-cell technologies has revealed the heterogeneity of microglia in both human and mouse, linking specific developmental, aging, and disease stages with distinct microglial states. Data analysis and integration with functional studies are essential for correlating genetic cues with the biological functions of microglia in order to dissect the 'microglial etiology' of aging and complex neurological diseases and identify therapeutic and diagnostic targets.

Restoring metabolism of myeloid cells reverses cognitive decline in ageing

Paras S. Minhas et al.

Summary: Ageing is characterized by chronic pro-inflammatory responses that affect various aspects of health, including the brain. The study shows that in ageing mice, myeloid cell bioenergetics are suppressed in response to increased signalling by the lipid messenger prostaglandin E-2 (PGE(2)), leading to maladaptive pro-inflammatory responses. By inhibiting myeloid EP2 signalling, it is possible to reverse cognitive ageing and restore youthful immune functions.

Astrocytes in stress accumulate lipid droplets

Tina Smolic et al.

Summary: The study found that the increase of LDs in astrocytes may be crucial for cell survival and proliferation. Under stress conditions, exposure to noradrenaline and metabolic stress strongly facilitated LD accumulation in astrocytes, potentially providing energy support and neuroprotection against stress-induced lipotoxicity.

Dystrophic microglia are associated with neurodegenerative disease and not healthy aging in the human brain

Ryan K. Shahidehpour et al.

Summary: The study found an age-associated increase in dystrophic microglia in the hippocampus and frontal cortex, but this increase was proportional to the total number of microglia, suggesting aging alone does not explain the presence of dystrophic microglia. Dystrophic microglia, rather than hypertrophic microglia, were found to be the disease-associated microglia morphology. Furthermore, changes in iron homeostasis in dystrophic microglia were identified as a potential molecular mechanism driving the degeneration of microglia in neurodegenerative disease.

Ensheathment and Myelination of Axons: Evolution of Glial Functions

Klaus-Armin Nave et al.

Summary: Myelination of axons is essential for rapid nerve conduction in vertebrates, with myelinating oligodendrocytes and Schwann cells playing additional roles in neuronal energy metabolism, similar to axon-ensheathing glial cells in invertebrates. Bidirectional axoglial interactions regulate cell fate and nerve conduction, and the evolution of compact myelin in vertebrates was a key step in adapting to new ecological niches.

Gut cytokines modulate olfaction through metabolic reprogramming of glia

Xiaoyu Tracy Cai et al.

Summary: Glial metabolic reprogramming by gut-derived cytokines in Drosophila results in lasting changes in the sensory system of an ageing organism. This metabolic reprogramming is transient in young flies but becomes constitutive in old flies, contributing to an age-related decline in sensory discrimination.

Glial AP1 is activated with aging and accelerated by traumatic brain injury

China N. N. Byrns et al.

Summary: The authors demonstrate that glial AP1 is initially protective after TBI but can lead to tau pathology and degeneration over time. This process may also occur with normal aging, suggesting that TBI could accelerate age-related degenerative processes.
