Carbon-centered radical capture at nickel(II) complexes: Spectroscopic evidence, rates, and selectivity

Qiao Lin et al.

Summary: This report presents an experimental characterization of the capture of carbon-centered radicals at nickel(II) centers in catalytic reactions. The study investigates various aspects, such as ligand structure-activity relationships, mechanism, kinetics, and stereoselectivity. Spectroscopic data supports the formation of a nickel(III) intermediate. The reactivity differences between nickel-aryl and nickel-alkyl complexes suggest different rate-determining steps for C(sp3)-C(sp3) and C(sp2)-C(sp3) bond formation. Kinetic data show capture rates on the scale of 107 M-1s-1 and 106 M-1s-1 for primary and secondary radicals, respectively. The activation energy is found to be higher than previously estimated through computational methods. Stoichiometric experiments with chiral nickel complexes demonstrate that the radical trapping step can confer diastereoselectivity and enantioselectivity with a significant ligand effect.

Site-Selective C(sp3)-H Functionalizations Mediated by Hydrogen Atom Transfer Reactions via α-Amino/α-Amido Radicals

Jaspreet Kaur et al.

Summary: Amines and amides, as nitrogen-containing compounds, are widely present in pharmaceuticals, natural products, agrochemicals, and peptides. Modifying the structure of amines and amides, especially at the nitrogen position, can significantly change their biological activity and improve the pharmacological properties of small drug molecules. Recent developments in hydrogen atom transfer (HAT) strategies have proven to be powerful in selectively activating amines and amides and facilitating the formation of C-C and C-X (X = heteroatom) bonds. Understanding the selective activation of C(sp(3))-H bonds and controlling reactivity is crucial for late-stage functionalization.

Visible-Light-Induced?,?-C(sp3)-H Difunctionalization ofPiperidines

Biao Wang et al.

Summary: Here, we present a novel protocol for visible-light-induced α,γ-C(sp3)-H difunctionalization of piperidines. This redox-neutral, atom-economical protocol, which has a wide substrate scope and good functional group compatibility, offers a concise and practical method for constructing piperidine-containing bridged-ring molecules. Preliminary mechanistic studies indicate that the highly regioselective activation of the inert γ-C(sp3)-H bond of piperidines is achieved through a 1,5-hydrogen atom transfer reaction of a nitrogen radical generated in situ.
