Mitochondria-Targeting Phototheranostics by Aggregation-Induced NIR-II Emission Luminogens: Modulating Intramolecular Motion by Electron Acceptor Engineering for Multi-Modal Synergistic Therapy

Tianfu Zhang et al.

Summary: Phototheranostic agents, integrating sensitive in situ fluorescence imaging and effective multi-model synergistic therapy, have emerged as promising tools for cancer theranostics. However, balancing the competitive radiative and nonradiative processes in photon energy transfer remains challenging. In this study, three simple molecules with electron donating-accepting structures were developed. The introduction of an acridinium unit as an electron acceptor led to the formation of TPEDCAc with aggregation-induced second near-infrared emission, high reactive oxygen species generation capability, and excellent photothermal conversion efficiency. The key factors for achieving NIR-II emission and balanced photodynamic/photothermal conversion were found to be large torsional angle and strong electron-withdrawing ability of the acridinium unit. Impressively, TPEDCAc exhibited mitochondria-targeting capability and high performance in in vivo multi-modal cancer theranostics under NIR laser irradiation. This work not only provides a single NIR-II AIE-based multi-modal cancer theranostic system but also inspires new insights into the development of theranostic platforms in the future.

Multifunctional liquid metal-based nanoparticles with glycolysis and mitochondrial metabolism inhibition for tumor photothermal therapy

Xing-Lan Ding et al.

Summary: This study designed a tumor acidity-responsive nanoparticle that combines ATP generation inhibition and photothermal therapy to enhance tumor treatment. By inhibiting glycolysis and increasing oxidative stress, the nanoparticles degrade and cause mitochondrial dysfunction, leading to reduced ATP production and heat shock protein expression, ultimately improving the therapeutic effect of photothermal therapy.

ER-Targeting Cyanine Dye as an NIR Photoinducer to Efficiently Trigger Photoimmunogenic Cancer Cell Death

He Ma et al.

Summary: This study developed the first ER-localizable immunogenic cell death (ICD) photoinducer that induces ER stress under mild near-infrared (NIR) irradiation using a thio-pentamethine cyanine dye (TCyS). The photoinducer has superior ROS generation capability in both normoxia and hypoxia conditions and leads to the production of damage-associated molecular patterns. In vivo experiments demonstrated its effectiveness in tumor treatment.

Strategies for efficient photothermal therapy at mild temperatures: Progresses and challenges

Peng Gao et al.

Summary: Photothermal therapy (PTT) is a method of eliminating tumors through hyperthermia induced by photothermal agents (PTAs) under laser irradiation. However, current methods require long, frequent, and high-power density laser irradiation, which can cause damage to the skin and healthy tissues. In contrast, low-temperature photothermal therapy (LTPTT) can inhibit tumors through mild hyperthermia, making it more attractive and feasible. This article systematically summarizes the strategies for achieving LTPTT, introduces various organic and inorganic materials used as PTAs, and discusses the challenges and future perspectives in this field.

Evoking tumor associated macrophages by mitochondria-targeted magnetothermal immunogenic cell death for cancer immunotherapy

Han Jiang et al.

Summary: This study proposes a novel immunogenic cell death (ICD) modality based on mitochondrial heat stress by magnetic hyperthermia treatment (MHT), which effectively evokes tumor-associated macrophages (TAMs) against cancer cells. A nanomedicine named MRT is synthesized for mitochondria-targeted heat stress-induced oxidative damage of tumor cells, leading to the release of damage-associated molecular patterns (DAMPs) and the activation of TAMs-mediated immune response against cancer cells.
