Aggregation Effect on Multiperformance Improvement in Aryl-Armed Phenazine-Based Emitters

Summary: This paper presents the first report of a new class of multifunctional aggregation-induced emission (AIE) luminogens based on 5,10-diarylphenazine (DPZ) derivatives with full-wavelength emission. Multiple properties, such as fluorescence intensity and free radical and type I reactive oxygen species (ROS) efficiencies, could be simultaneously activated from the unimolecular level to the aggregate state. The mechanisms of this multiple performance improvement are discussed in detail based on sufficient performance characterization, and some of the newly prepared AIEgens exhibited toxicity to cancer cells during photodynamic therapy.

Organic Persistent RTP Crystals: From Brittle to Flexible by Tunable Self-Partitioned Molecular Packing

Summary: Organic RTP crystals with elastic/plastic deformation are achieved by controlling molecular configurations. These crystals have divisional aggregation structures of aromatics and alkoxy chains, and the RTP property can be modulated. The highest record of RTP lifetime is 972.3 ms for organic flexible crystals. The relationship among elastic/plastic deformation, RTP property, and aggregated structures is established, and a new approach for the development of flexible RTP crystals is explored.

pH-Triggered Charge Reversible Fluorescent Probe for Simultaneous Imaging of Lipid Droplets and Nucleoli in Living Cells

Summary: Cooperation between organelles, such as lipid droplets (LDs) and nucleoli, is crucial for maintaining normal cellular functions. However, there has been limited in situ observation of the interaction between them due to the lack of suitable tools. In this study, a pH-triggered charge reversible fluorescent probe (LD-Nu) was developed to visualize the physical contact between LDs and nucleoli for the first time. Cell imaging results showed that LDs in both the cytoplasm and nucleus were susceptible to external stimuli, with cytoplasmic LDs being more affected. LD-Nu can serve as a powerful tool for further investigating the interaction mechanism between LDs and nucleoli in living cells.

In Vivo Imaging of Lipid Droplets and Oxygen Status in Hepatic Tissues of Nonalcoholic Fatty Liver Model Mice Using a Lipophilic Ir(III) Complex

Summary: Nonalcoholic fatty liver disease (NAFLD) is a commonly occurring condition worldwide. In this study, the phosphorescent Ir(III) complex BTP was developed as the first probe that can visualize lipid droplets (LDs) and oxygen status in liver tissues simultaneously. BTP was efficiently taken up by adipocytes and selectively accumulated in LDs. Quantifying oxygen levels in LDs allowed for tracking of cellular oxygen tension changes. In mice, BTP enabled specific visualization of LDs in hepatic lobules and imaging of the oxygen gradient. NAFL model mice fed a high-fat diet showed increased LDs and reduced oxygen tension, leading to microvascular structural abnormalities and partial hypoxia.

Solvatochromic Near-Infrared Aggregation-Induced Emission-Active Acrylonitriles by Acceptor Modulation for Low-Power Stimulated Emission Depletion Nanoscopy

Summary: A series of donor-acceptor-acceptor structured fluorescent acrylonitriles featuring aggregation-induced emission (AIE) were developed for low depletion power STED super-resolution imaging. These materials exhibited tunable near-infrared emissions, high fluorescence quantum yields, significant Stokes shifts, and large two-photon absorption cross-sections. They also showed solvatochromic effect and ultrahigh fluorescence quantum yields in low-polarity solvents.

Unraveling Molecular Assembly and Tracking Lipid Droplet Dynamics Using Fluorescent Phenanthroimidazole Derivatives

Summary: This study unraveled the real-time dynamic molecular self-assembly processes and reversible morphological transformation between supramolecular fibers and spherical nanoaggregates of alkyl chain-substituted phenanthroimidazole-based luminogen (BPIB1) using fluorescence lifetime imaging microscopy (FLIM). The presence of an octyl chain and basic nitrogen centers in BPIB1 led to distinct self-assembly patterns. The smaller-sized fluorescent nanoaggregates of BPIB1 were used to track the dynamics of lipid droplets in live cells and C. elegans, demonstrating new avenues to explore the formation pathways of molecular aggregates and complex organelle dynamics.

Ultrabright and Highly Polarity-Sensitive NIR-I/NIR-II Fluorophores for the Tracking of Lipid Droplets and Staging of Fatty Liver Disease

Summary: This study developed two bright NIR-I/NIR-II fluorophores with D-pi-A-pi-D structures for tracking fatty liver disease progression in vivo. These fluorophores exhibited ultrahigh quantum yields and ultrasensitive fluorescence response to environment polarity variations. By using these fluorophores as fluorescent nanoprobes, high-resolution imaging of vessels and accurate identification and imaging-guided surgeries of submillimeter scale lymph nodes were successfully achieved. Additionally, the fluorophores showed superior performance in indicating lipid droplets in living cells, enabling precise monitoring and staging of fatty liver disease with a contact-free method utilizing NIR-I/NIR-II fluorescent LDs probe.

Exploiting the Twisted Intramolecular Charge Transfer Effect to Construct a Wash-Free Solvatochromic Fluorescent Lipid Droplet Probe for Fatty Liver Disease Diagnosis

ANI showed enhanced fluorescence in low-polar environments, allowing for selective lighting up of lipid droplets with high contrast. Efficiently emissive, strongly solvatochromic and lipid droplet-specific, fluorescent probes for mapping polarity in vitro

Summary: In this study, two isoindole-based fluorescent probes were synthesized with solvatochromic effects, large two-photon absorption cross-section, specificity, high photostability, and excellent performance in monitoring lipid droplet polarity changes in living cells. These probes exhibited high sensitivity between emission wavelength maxima and solvent polarity due to the existence of intramolecular charge transfer process. They also maintained high fluorescence quantum yields. The probes were successfully used for detecting lipid droplet polarity in living cells by confocal laser scanning fluorescence imaging. Importantly, the probes demonstrated photostability after degradation in a zebrafish model, making them suitable for long-term tracking and observation of biological dynamic changes.
