Template-Directed Synthesis of Bivalent, Broad-Spectrum Hosts for Neuromuscular Blocking Agents**

Allison J. Selinger et al.

Summary: This study reports the synthesis of bivalent water-soluble calix[4]arene and calix[5]arene hosts, Super-sCx4 and Super-sCx5, as new broad-spectrum supramolecular binders of neuromuscular blocking agents. The hosts bind a variety of NMBAs by engaging both quaternary amines present, with high selectivity over other hydrophobic amines. This binding is achieved through linking two highly anionic p-sulfonatocalixarene building blocks in aqueous solution using the target bisquaternary amine NMBAs as a template.

Combining Excellent Selectivity with Broad Target Scope: Biosensing with Arrayed Deep Cavitand Hosts

Wenwan Zhong et al.

Summary: Water-soluble, self-folding deep cavitands can overcome the trade-off between selectivity and scope in traditional macrocyclic host-based biosensing. These hosts have flexible structures and large cavities that can bind to various fluorescent dyes, causing fluorescence enhancement or quenching upon binding. By utilizing the affinity of the hosts for NMe3+ groups, site-selective recognition of post-translational lysine methylations can be achieved. Additionally, these hosts can selectively recognize serine phosphorylation, lysine acetylation, and arginine citrullination through non-cavity-based interactions with charged biomolecules.

Kenichi Kato et al.

Summary: Pillar[n]arenes are pillar-shaped macrocyclic compounds with unique structures and excellent properties, such as versatility, substituent-dependent solubility, cavity-size-dependent host-guest properties, and unit rotation. These advantages enable the high-yield synthesis and rational design of pillar[n]arene-based mechanically interlocked molecules (MIMs) that can dynamically convert between interlocked and unlocked states. The highly symmetrical pillar-shaped structures of pillar[n]arenes are also useful for studying the motion of pillar[n]arene wheels in MIMs and creating sophisticated MIMs with higher-order structures.
