The NADPARK study: A randomized phase I trial of nicotinamide riboside supplementation in Parkinson's disease

Brage Brakedal et al.

Summary: In this study, we found that oral intake of nicotinamide riboside (NR) in Parkinson's disease (PD) patients was safe and increased cerebral NAD levels, leading to altered cerebral metabolism and mild clinical improvement. NR also upregulated processes related to mitochondrial, lysosomal, and proteasomal function in blood cells and/or skeletal muscle, and decreased the levels of inflammatory cytokines.

Epigenome-wide association study of human frontal cortex identifies differential methylation in Lewy body pathology

Lasse Pihlstrom et al.

Summary: In this study, the authors investigated the role of DNA methylation changes in the pathogenesis of Parkinson's disease (PD) and dementia with Lewy bodies (DLB). They identified differentially methylated genomic loci associated with Lewy pathology, providing insights into novel disease pathways and potential molecular studies of Lewy body pathology.

Neuropathological consensus criteria for the evaluation of Lewy pathology in post-mortem brains: a multi-centre study

Johannes Attems et al.

Summary: Several staging systems exist for the neuropathological diagnosis of LBD, with varying levels of inter-rater reliability. The LPC system, based on a dichotomous approach for scoring LP, showed good reproducibility and allowed for the classification of all cases into distinct categories. It may serve as a reliable and useful standard future approach for the post-mortem evaluation of LP.

Genome-wide histone acetylation analysis reveals altered transcriptional regulation in the Parkinson's disease brain

Lilah Toker et al.

Summary: Parkinson's disease is a complex neurodegenerative disorder associated with mitochondrial dysfunction and epigenetic dysregulation. Research found increased histone acetylation levels in PD, particularly at the H3K27 site, and a decoupling between H3K27 acetylation and gene expression in the disease, especially among nuclear-encoded mitochondrial genes. This suggests aberrant histone acetylation and altered transcriptional regulation play a role in the pathophysiology of PD.

Summary: Mitochondria play a significant role in age-related neurodegenerative diseases, with complex involvement and interactions with numerous genes and proteins. Understanding the regulation of mitochondrial pathways may be beneficial in finding interventions to delay the onset of neurodegenerative diseases.

Cross-platform transcriptional profiling identifies common and distinct molecular pathologies in Lewy body diseases

Rahel Feleke et al.

Summary: Using paired bulk-tissue and single-nucleus RNA-sequencing, researchers identified transcriptional alterations in multiple cell types across PD, PDD, and DLB, with evidence for widespread dysregulation of RNA splicing particularly in PDD and DLB. Potential splicing factors were identified, linking to other dementia-related neurodegenerative diseases, coordinating this dysregulation, and transcriptomic commonalities and distinctions between the LBDs were elucidated, informing the relationships between these three clinical disorders.

Summary: Recent advances in RNA sequencing technologies have revealed the significance of long non-coding RNAs (lncRNAs) in modulating protein-coding gene expression, especially in the central nervous system. Therefore, lncRNAs are gaining attention as potential novel therapeutic targets for brain disorders.
