Altered SOD1 maturation and post-translational modification in amyotrophic lateral sclerosis spinal cord

Benjamin G. Trist et al.

Summary: This study examined the changes in SOD1 protein in post-mortem spinal cord tissues of ALS patients. The results showed mislocalization and accumulation of SOD1 protein in motor neurons of ALS patients, which was associated with instability and mismetallation of enzymatically active SOD1 dimers, as well as alterations to SOD1 post-translational modifications and molecular chaperones governing SOD1 maturation. These changes mostly occurred in regions of neurodegeneration and differentiated ALS patients from controls effectively.

De novo mutations in SOD1 are a cause of ALS

Kathrin Muller et al.

Summary: De novo mutations in SOD1 have been identified as a cause of sporadic ALS, potentially impacting both isolated cases and smaller familial groups. While the exact origin of these mutations remains uncertain, the findings suggest the importance of genetic counseling and screening for all ALS patients to potentially benefit from personalized therapy.

Metal migration and subunit swapping in ALS-linked SOD1 Zn2+ transfer between mutant and wild-type occurs faster than the rate of heterodimerization

Chad M. Dashnaw et al.

Summary: This study investigated heterodimerization and metal transfer between WT SOD1, ALS-variant SOD1, and deamidated SOD1. The rates and free energies of heterodimerization were determined, and it was found that partially metalated heterodimers have similar properties to fully metalated heterodimers. The study also demonstrated that zinc-replete SOD1 can transfer Zn2+ to apo-SOD1, and suggested that WT apo-SOD1 may increase the toxicity of mutant SOD1 by stealing its zinc.

Amyotrophic lateral sclerosis

Eva L. Feldman et al.

Summary: Amyotrophic lateral sclerosis, a fatal CNS neurodegenerative disease, lacks optimal management strategies. Improved understanding of clinical presentation, pathophysiology, and diagnostic criteria has not yet translated into direct patient benefit. However, the concept of amyotrophic lateral sclerosis as a complex syndrome is already influencing clinical practice.

Summary: This study conducted whole exome sequencing to analyze SOD1 mutations in Chinese ALS patients, identifying two novel variants and summarizing the natural history of SOD1-mutated patients. It was found that male patients had better survival, and familial ALS patients received earlier diagnosis.

Redox metabolism: ROS as specific molecular regulators of cell signaling and function

Claudia Lennicke et al.

Summary: Redox reactions play a crucial role in energy metabolism and physiological functions of organisms. Reactive oxygen species (ROS), such as hydrogen peroxide, are important derivatives in normal aerobic metabolism. Hydrogen peroxide can modify protein activity by oxidizing cysteine residues, affecting enzyme and transcription factor functions and cellular processes.

Free energy calculations of amyotrophic lateral sclerosis-causing superoxide dismutase 1 mutants reveal common perturbations to stability and dynamics along the maturation pathway

Nicholas G. M. Wells et al.

Summary: SOD1 has been a key target in ALS research, but mutations have a significant impact on free energy changes during maturation, highlighting the need for state-specific therapeutic targeting. Many mutations show a disrupted allosteric network within SOD1, which may contribute to explaining SOD1-linked ALS disease.
