Hybrid molecules combining GABA-A and serotonin 5-HT6 receptors activity designed to tackle neuroinflammation associated with depression

Summary: There is evidence showing that the combination of GABA molecule and chemical fragments interacting with the serotonin 5-HT6 receptor can curb neuroinflammation and provide antidepressant-like activity. The hybrid molecule 3B has shown promising affinity for 5-HT6 receptors and agonistic properties on GABA-A receptors. In vivo studies have demonstrated its robust antidepressant-like activity and effectiveness in reducing oxidative stress markers and the pro-inflammatory cytokine IL-6.

GPCRs steer Gi and Gs selectivity via TM5-TM6 switches as revealed by structures of serotonin receptors

Summary: This study reveals the mechanism of selectivity in coupling between serotonin receptors and G proteins. It is found that the length of TM5 and TM6 can act as a macro-switch to determine the receptor's preference for G(s) or G(i) proteins. Additionally, specific residues within TM5 and TM6 function as micro-switches to form specific interactions with G(s) or G(i). These findings extend our understanding of ligand recognition at serotonin receptors.
