Discovery of novel dual acetylcholinesterase and butyrylcholinesterase inhibitors using machine learning and structure-based drug design

Manish Kumar Tripathi et al.

Summary: This study identified a novel dual acetylcholinesterase and butyrylcholinesterase inhibitor using a multistep virtual screening method combined with machine learning models and structural bioinformatics. The compound CD05692 showed potential inhibitory effects against AChE and BChE enzymes and could be further optimized as a lead for designing potential cholinesterase inhibitors to combat Alzheimer's disease.

Modulation of KIF17/NR2B crosstalk by tozasertib attenuates inflammatory pain in rats

Ankit Uniyal et al.

Summary: Chronic pain is a burdensome disorder affecting millions of people worldwide, and recent studies have revealed the important role of kinesin nanomotors. In an animal model, the aurora kinase inhibitor tozasertib can alleviate acute inflammatory pain and suppress hypersensitivity to different stimuli. Further investigations demonstrate that tozasertib can also attenuate oxidative stress and immune cell activation induced by chronic pain. These findings suggest that tozasertib exerts its anti-nociceptive effects by inhibiting the aurora kinase-mediated signaling.

Neuroprotective Effect of Epalrestat on Hydrogen Peroxide-Induced Neurodegeneration in SH-SY5Y Cellular Model

Sivakumar Lingappa et al.

Summary: Epalrestat (EPS) is an aldose reductase inhibitor that penetrates the blood-brain barrier and is used in the treatment of diabetic neuropathy. It has been found to reduce neurodegeneration by inhibiting ROS-induced oxidative damage and has shown potential in regulating Alzheimer's disease pathology through GSK3 beta and tau protein phosphorylation. An optimal concentration of EPS therapy may decrease AD progression by reducing tau phosphorylation levels.
