Facile Synthesis of Clickable Unnatural Sugars in the Unprotected and 1,6-Di-O-Acylated Forms for Metabolic Glycan Labeling

Bo Cheng et al.

Summary: Clickable unnatural sugars are widely used in studying glycosylation in living systems. The protection of unnatural sugars by 1,6-di-O-acylation improves labelling efficiency and avoids non-specific S-glyco-modification. In this study, a series of clickable unnatural sugars were synthesized in both protected and unprotected forms, and the 1,6-di-O-propionylated unnatural sugars were found to be optimal for glycan labelling. This work provides a convenient synthetic route for investigating the functional roles of glycans without artificial S-glyco-modification.

Metabolic Glycoengineering with Azide- and Alkene-Modified Hexosamines: Quantification of Sialic Acid Levels

Jeremias E. G. A. Dold et al.

Summary: Metabolic glycoengineering (MGE) is a method utilized to incorporate chemical reporter groups into cellular glycans, allowing for visualization and isolation of glycans and potentially targeting drugs to cancer cells. The study introduced a new labeling reaction using 1,2-diamino-4,5-methylenedioxybenzene for quantification of sialic acid derivates after MGE. It also explored the effects of various ManNAc derivatives on sialic acid production in different cell types.

Glycan Labeling and Analysis in Cells and In Vivo

Bo Cheng et al.

Summary: Glycans, as one of the major biomacromolecules in cells, are crucial in various biological processes. Recent advances in labeling, imaging, and profiling of glycans have enabled researchers to better understand their biological functions in physiological or pathological contexts. These methods and strategies have facilitated the visualization, tracking, and enrichment of glycans, as well as the selective labeling and imaging of glycans in cells and tissues.
