Modular Halogenation, α-Hydroxylation, and Acylation by a Remarkably Versatile Polyketide Synthase

Franziska Hemmerling et al.

Summary: In this study, a remarkably versatile trans-acyltransferase (trans-AT) PKS from Serratia was characterized, which builds structurally complex macrolides via over ten functionally distinct PKS modules. A new oxygenation module and a halogenating module were identified, along with modular O-acetylation.

Poly(dehydroalanine): Synthesis, Properties, and Functional Diversification of a Fluorescent Polypeptide

Isaac Benavides et al.

Summary: A new soluble poly(S-alkyl-L-cysteine) precursor was designed for the successful preparation of long-chain poly(dehydroalanine) and the incorporation of dehydroalanine residues and A(DH) segments into copolypeptides. A(DH) was found to adopt a unique hybrid coil structure and possess inherent blue fluorescence suitable for imaging applications. Its reactive electrophilic groups allow for efficient modification under mild conditions, making it a unique component for self-assembled biomaterials.

Tackling the emerging threat of antifungal resistance to human health

Matthew C. Fisher et al.

Summary: This review emphasizes the important threat of invasive fungal infections to public health and the emergence of antifungal resistance. It discusses the main mechanisms of antifungal resistance and compares them with bacterial resistance to antimicrobial control. The review also explores the research and innovation topics needed to minimize the occurrence and impact of antifungal resistance, including environmental links, surveillance, diagnostics, transmission routes, novel therapeutics, and hotspot mitigation strategies. The need for global efforts to manage existing antifungal drugs and direct future research and development of therapies and interventions is highlighted.

Structure of a Promiscuous Thioesterase Domain Responsible for Branching Acylation in Polyketide Biosynthesis

Amy E. Fraley et al.

Summary: This study provides the first structural insights into a distinct group of TEs (TE(B)s) that have diverse acylation roles in polyketide and peptide biosynthesis. By analyzing and engineering the active site, the acyl-group acceptance can be modulated. Moreover, the production of longer chain branched oocydins was discovered, positioning this enzyme as a valuable synthetic biology tool for polyketide drug development.

Solanimycin: Biosynthesis and Distribution of a New Antifungal Antibiotic Regulated by Two Quorum-Sensing Systems

Miguel A. Matilla et al.

Summary: The emergence of drug-resistant fungal infections has led to the search for new compounds to combat fungal pathogens. This study describes the discovery and characterization of a new antifungal compound, solanimycin, produced by a hybrid polyketide/nonribosomal peptide system. Solanimycin exhibited broad-spectrum antifungal activity against plant-pathogenic fungi and Candida albicans. The genomic cluster responsible for solanimycin production was identified and analyzed, and its production was enhanced under certain conditions. The study suggests potential utility of solanimycin for agricultural and clinical exploitation.

Discovery and Biosynthetic Investigation of a New Antibacterial Dehydrated Non-Ribosomal Tripeptide

Shan Wang et al.

Summary: This study discovered a new antimicrobial peptide, albopeptide, containing two contiguous unsaturated residues, Dha-(E)-Dhb, with narrow-spectrum activity against vancomycin-resistant Enterococcus faecium. The research also demonstrated that albopeptide is synthesized through a noncanonical NRPS pathway and revealed the presence of a previously untapped group of short unsaturated peptides in the bacterial kingdom, suggesting an important biological function in bacteria.
