Antibody-drug conjugates: Smart chemotherapy delivery across tumor histologies

Paolo Tarantino et al.

Summary: Recent years have seen a shift in cancer treatment towards therapies tailored to specific molecular aberrations and immunologic markers, rather than solely relying on histology. Several histology-agnostic therapies are now adopted in clinical practice for treating patients, and novel antibody-drug conjugates (ADCs) have shown promise in treating solid tumors. The multihistological expansion of ADCs poses both challenges and opportunities for cancer treatment.

Leonurine alleviates ferroptosis in cisplatin-induced acute kidney injury by activating the Nrf2 signalling pathway

Jianqiang Hu et al.

Summary: Evidence suggests that Nrf2 activation has renal protective effects on cisplatin-induced acute kidney injury, potentially by inhibiting lipid peroxide-mediated ferroptosis. Leonurine activates the Nrf2 pathway and protects against cisplatin-induced acute kidney injury by inhibiting ferroptosis.

Targeting iron metabolism using gallium nanoparticles to suppress ferroptosis and effectively mitigate acute kidney injury

Xishao Xie et al.

Summary: This study synthesized gallic acid-gallium polyvinyl pyrrolidone nanoparticles (GGP NPs) as a potential iron-scavenging agent and demonstrated their good biocompatibility and protective effects in inhibiting ferroptosis. Treatment with GGP NPs significantly ameliorated renal injury and mitochondrial damage, making them a potential candidate for AKI treatment.

Ultrasmall Folate Receptor Alpha Targeted Enzymatically Cleavable Silica Nanoparticle Drug Conjugates Augment Penetration and Therapeutic Efficacy in Models of Cancer

Fei Wu et al.

Summary: An ultrasmall folate receptor alpha (FR alpha) targeted silica nanoparticle (C'Dot) drug conjugate (CDC) was developed to address the limitations of current systemic drug delivery for cancer management. The lead candidate formulation demonstrated specific targeting, multivalent binding properties, enzymatic responsive payload cleavage, stability in human plasma, rapid clearance, and minimal organ retention. It also showed superior efficacy in tumor penetration, cytotoxicity, and therapeutic outcome in various tumor models expressing different levels of FR alpha.

Nuciferine protects against folic acid-induced acute kidney injury by inhibiting ferroptosis

Danyu Li et al.

Summary: Nuciferine effectively mitigates pathological changes and kidney dysfunction in mice with folic acid-induced acute kidney injury, as well as prevents RSL3-induced ferroptotic cell death in vitro. The protective effect of nuciferine against ferroptosis is associated with its ability to prevent iron accumulation and lipid peroxidation, as well as its interaction with GPX4.
