Broad lipid phase transitions in mammalian cell membranes measured by Laurdan fluorescence spectroscopy

Summary: By using fluorescence spectroscopy and the membrane-embedded dye Laurdan, it was experimentally shown that linear changes of cell membrane order in the physiological temperature regime are part of broad order-disorder-phase transitions. The study also revealed that membrane order changes are sensitive to factors such as cholesterol content, pH, anaesthetics, as well as temperature changes, adaptation to culture conditions, and morphological changes. The observed changes in membrane order were found to be equivalent to temperature changes in the range of +12 K < dT < -6 K.

The living state: How cellular excitability is controlled by the thermodynamic state of the membrane

Experimental data and literature review predict parameter ranges for membrane transitions, explaining changes in action potential velocity, loss of excitability, and triggers for action potentials. This study suggests that the thermodynamic properties of membrane interfaces, rather than specific molecules, control excitability in living systems.
