Nonmalignant portal vein thrombi in patients with cirrhosis consist of intimal fibrosis with or without a fibrin-rich thrombus

Ellen G. Driever et al.

Summary: The study found that portal vein thrombosis in cirrhotic patients is mainly composed of intimal fibrosis, with fibrin-rich thrombus present in only one-third of cases. Clinical characteristics showed no significant association with the presence of fibrin-rich thrombi.

Fibrin clot quality in acutely ill cirrhosis patients: Relation with outcome and improvement with coagulation factor concentrates

Annabel Blasi et al.

Summary: The study found that patients with AD and ACLF had higher thrombin generation but lower levels of prothrombin, fibrinogen, and factor XIII compared to healthy controls. Fibrinogen concentrate improved clot quality, while prothrombin complex concentrate increased clot resistance. Further research is needed to determine the clinical benefits of coagulation factor concentrates in preventing or treating bleeding in liver disease patients.

Effects of Inflammation on Hemostasis in Acutely Ill Patients with Liver Disease

Ellen G. Driever et al.

Summary: Patients with liver diseases experience imbalances in their hemostasis, which can be further destabilized by disease complications. Systemic inflammation in these patients leads to alterations in the hemostatic system, potentially causing local ischemia, liver disease progression, and multiorgan failure. Anti-inflammatory treatment may prove beneficial in preventing thrombotic or bleeding complications and halting liver disease progression.

Citrullinated fibrinogen forms densely packed clots with decreased permeability

Summary: The effect of fibrinogen citrullination on the structure of fibrin clots is investigated in this study. The results show that citrullination leads to thinner, denser, and less porous clots.

Transfusion with Cryoprecipitate for Very Low Fibrinogen Levels Does Not Affect Bleeding or Survival in Critically Ill Cirrhosis Patients

Isadore M. Budnick et al.

Summary: Low fibrinogen levels in cirrhosis patients may reflect severity of illness, but are not directly associated with 30-day mortality. Transfusion of cryoprecipitate to treat low fibrinogen levels does not independently affect survival or bleeding complications.

Factor XIII cross-links fibrin(ogen) independent of fibrin polymerization in experimental acute liver injury

Lauren G. Poole et al.

Summary: This study investigated the mechanisms of fibrin polymerization and cross-linking in acute liver injury induced by acetaminophen overdose. It was found that deficiency in FXIII dramatically reduced fibrin(ogen) cross-linking, while the impact on fibrin(ogen) deposition and cross-linking was less significant in Fib(AEK) mice. The presence of oxidative mediators in the injured liver was proposed as a factor affecting fibrin polymerization without affecting FXIII-mediated cross-linking.

PAD4 takes charge during neutrophil activation: Impact of PAD4 mediated NET formation on immune-mediated disease

Xiaosong Liu et al.

Summary: This review provides insights into the structure-activity relationships of PAD4 and its involvement in autoimmune disorders and thrombo-inflammatory diseases, highlighting the crucial role of PAD4-mediated NET formation in the pathogenesis of various diseases. Advances in understanding PAD4 function are essential for the development of specific inhibitors to combat and prevent autoimmune disorders and inflammatory diseases.

Fibrinolysis in Patients with Liver Disease

Fien A. von Meijenfeldt et al.

Summary: Patients with liver disease exhibit complex changes in their hemostatic system, with debates on the presence of hyperfibrinolysis and its association with bleeding risk. The lack of clear definitions and reliable tests for hypo- and hyperfibrinolysis in liver disease complicates the understanding of their fibrinolytic state. Large clinical studies on the relationship between bleeding and fibrinolysis in patients with liver disease are limited, highlighting the need for further research in this area.

Thrombus Structural Composition in Cardiovascular Disease

Ghadir Alkarithi et al.

Summary: Thrombosis is a major complication of cardiovascular disease, with thrombi composed of fibrin, red blood cells, platelets, leukocytes, and neutrophil extracellular traps. Different diseases exhibit thrombi with varying compositions, highlighting the importance of further research on thrombus composition and function to enhance treatment and clinical outcomes.

Elimination of fibrin γ-chain cross-linking by FXIIIa increases pulmonary embolism arising from murine inferior vena cava thrombi

Cedric Duval et al.

Summary: This study demonstrated that the lack of fibrin gamma-chain cross-linking leads to weakened thrombi that are more prone to fragmentation, increasing the risk of embolization. The findings also highlight the important role of fibrin gamma-chain cross-linking in clot stability and provide mechanistic targets for preventing thrombosis through selective modulation of activated Factor XIII-mediated fibrin alpha-chain cross-linking.
