Measurement of autophagic flux in humans: an optimized method for blood samples

Julien Bensalem et al.

Summary: Autophagic flux, a critical cellular process, can be increased by targeting nutrition-related biochemical signaling. This study introduces a method to directly measure human autophagic flux from blood samples by treating whole blood and using isolated cells to measure LC3B-II protein. The method is valuable for evaluating intra-individual variation induced by interventions.

Comparison of chloroquine-like molecules for lysosomal inhibition and measurement of autophagic flux in the brain

Celia Fourrier et al.

Summary: Measurement of autophagic flux in the brain remains a challenging research topic, with current methods not fully effective in capturing autophagic activity in this specific tissue. More research is needed to find a suitable drug for this purpose.

Human autophagy measurement: an underappreciated barrier to translation

Timothy J. Sargeant et al.

Summary: Preclinical research suggests that autophagy could be modified to prevent age-related diseases in humans. Despite this, the translation of this knowledge into clinical practice has not yet been achieved. Measuring human autophagy capacity is crucial for this translation process.

Inhibiting mTOR activity using AZD2014 increases autophagy in the mouse cerebral cortex

Julien Bensalem et al.

Summary: Inhibiting mTOR using AZD2014 was shown to increase autophagic activity in both the liver and brain, which could be valuable in addressing conditions associated with defective autophagy, especially neurodegenerative diseases.

Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)

Daniel J. Klionsky et al.

Summary: The guidelines presented in this article aim to assist researchers in selecting and interpreting methods to examine autophagy, as well as to help reviewers provide constructive critiques of reports focused on these processes. It emphasizes that no single assay is perfect for every situation, and multiple techniques may be needed to properly monitor autophagy in different experimental settings. The article also highlights the importance of targeting multiple autophagy-related genes in genetic approaches to block autophagy effectively.
