ERAP2 Inhibition Induces Cell-Surface Presentation by MOLT-4 Leukemia Cancer Cells of Many Novel and Potentially Antigenic Peptides

Ioannis Temponeras et al.

Summary: Recent studies have found that pharmacological inhibition of ER aminopeptidase 2 (ERAP2) could potentially improve the efficacy of cancer immunotherapy. In this study, MOLT-4 T lymphoblast leukemia cells were treated with a selective ERAP2 inhibitor, resulting in significant shifts in the immunopeptidome. The newly generated peptides induced by the inhibitor could trigger cytotoxic responses against cancer cells and enhance the therapeutic effect of immune-checkpoint inhibitors.

Inhibitor-Dependent Usage of the S1′ Specificity Pocket of ER Aminopeptidase 2

Anastasia Mpakali et al.

Summary: Endoplasmic reticulum aminopeptidase 2 (ERAP2) is an important enzyme involved in immune response regulation and has potential as a pharmacological target. This study solved the crystal structures of ERAP2 in complex with two inhibitors, revealing that the usage of the S1' pocket in ERAP2 is dependent on the inhibitor, providing useful templates for further drug optimization.

Ligand-Directed Modification of Active Matrix Metalloproteases: Activity-based Probes with no Photolabile Group

Monika Kaminska et al.

Summary: Activity-based probes can discriminate active enzymes from inactive counterparts, targeting metalloproteases through reversible inhibitors with photo-crosslinkers. Novel probes were developed to covalently modify active matrix metalloproteases without external trigger, allowing for nanogram-scale reactions with recombinant MMPs in complex proteomes. This ligand-directed chemistry successfully labeled active MMP-12 secreted by eukaryote cells, with potential applications for other metalloproteases in unresolved proteomic profiling in vivo.
