Lung-selective mRNA delivery of synthetic lipid nanoparticles for the treatment of pulmonary lymphangioleiomyomatosis

Min Qiu et al.

Summary: The challenge of delivering mRNA to specific organs and cells in vivo is being addressed, with research showing that N-series LNPs can selectively deliver mRNA to the lungs while O-series LNPs tend to deliver mRNA to the liver. By analyzing the protein corona on targeted LNPs, unique plasma proteins were identified that contribute to their targetability. This research demonstrates the potential of LNPs as a promising therapeutic intervention for lung diseases like lymphangioleiomyomatosis.

Extracellular Vesicles: Interplay with the Extracellular Matrix and Modulated Cell Responses

Aleen Al Halawani et al.

Summary: The discovery of extracellular vesicles (EVs) carrying regulatory proteins, nucleic acids, and lipids has provided insights into how cells communicate and function. The bioactivity of EVs depends on their contents and surface molecules, and their interaction with the extracellular matrix (ECM) plays a role in tissue maintenance and repair. EVs can modulate matrix deposition and degradation, and contribute to tissue repair and regeneration.

Spermine Modified PNIPAAm Nano-Hydrogel Serving as Thermo-Responsive System for Delivery of Cisplatin

Soheila Ghasemi et al.

Summary: PNIPAAm hydrogel was synthesized using RAFT polymerization and modified with a pH-sensitive linker (spermine) to conjugate the anti-tumor drug, cisplatin. The drug release from this system showed temperature and pH-dependent behavior, with increased release at the polymer's transition temperature. In vitro toxicity assessment demonstrated the efficient killing of breast cancer cells.

Tuning Polymer Hydrophilicity to Regulate Gel Mechanics and Encapsulated Cell Morphology

Renato S. Navarro et al.

Summary: Mechanically tunable hydrogels are attractive platforms for 3D cell culture, and the stiffness of the hydrogel can be controlled by tuning the hydrophilicity of a protein. By bioconjugating the protein with azide-containing molecules of increasing hydrophilicity, the transition temperature of the protein can be increased, allowing direct control of the gel stiffness. These hydrogels have tunable moduli within a range relevant to soft tissues and can affect cell viability and spreading.

Increasing Angiogenesis Factors in Hypoxic Diabetic Wound Conditions by siRNA Delivery: Additive Effect of LbL-Gold Nanocarriers and Desloratadine-Induced Lysosomal Escape

Elnaz Shaabani et al.

Summary: Impaired wound healing in people with diabetes is caused by multiple factors, with lack of new blood vessel formation being one of the most important. Stabilization of HIF-1 alpha by downregulating PHD-2 can upregulate pro-angiogenic factors, improving diabetic wound healing. Gold nanoparticles loaded with siRNA and coated with a Poly L-arginine outer layer showed enhanced endosomal escape and transfection efficiency compared to other formulations.
