Capsule carbohydrate structure determines virulence in Acinetobacter baumannii

Summary: The study found that a mobile DNA fragment in a capsule assembly gene led to the high virulence of Acinetobacter baumannii, while repairing the gene resulted in the virulent strain becoming avirulent, with a single sugar being crucial for innate immune cells recognition and phagocytosis. By comparing genomes of A. baumannii strains, a specific gene variation associated with virulence was identified, showing that loss of a single capsule assembly gene dramatically altered virulence by affecting interactions between bacteria and host immune system.

Protected N-Acetyl Muramic Acid Probes Improve Bacterial Peptidoglycan Incorporation via Metabolic Labeling

Summary: Metabolic glycan probes have been successfully incorporated into bacterial cell walls with the development of a scalable synthesis of peptidoglycan precursors. The masking of polar groups on the glycan probes increases their overall incorporation into bacterial cells, as shown through cellular assays. Bacterial esterases play a key role in removing the masking groups for peptidoglycan biosynthesis, leading to a 10-fold improvement in probe utilization. This study advances our understanding of bacterial cell wall biology and provides insights on optimizing the delivery and usage of bacterial metabolic glycan probes.
