Synthesis of novel glutarimide ligands for the E3 ligase substrate receptor Cereblon (CRBN): Investigation of their binding mode and antiproliferative effects against myeloma cell lines

Summary: In order to enhance the chemical toolkit for targeted protein degradation, the authors developed a new series of non-thalidomide Cereblon (CRBN) ligands. By using a thio-Michael addition reaction, readily available 2-methylidene glutarimide was converted to a series of 2-((hetero)aryl(methyl))thio glutarimides. These compounds were evaluated for their affinity to human CRBN and their binding modes were studied through X-ray crystallography. The newly identified Cereblon ligands show promising potential for the synthesis of novel PROTAC protein degraders.

E3 ligase ligand chemistries: from building blocks to protein degraders

Summary: Proteolysis-targeting chimeras (PROTACs) have shown great therapeutic potential and usefulness as molecular biology tools in recent years. These compounds induce targeted protein degradation by forming a ternary complex and leading to E3 ligase-mediated ubiquitination and proteasomal degradation. The field of PROTAC has grown substantially due to advancements in the discovery of non-peptidic E3 ligase ligands. Expanding the range of E3 ligands could contribute to broadening the scope of targeted protein degradation. This review focuses on the chemistry aspect of PROTAC design and serves as a blueprint for future research in this complex field.

Development of MDM2 degraders based on ligands derived from Ugi reactions: Lessons and discoveries

Summary: PROTACs have attracted significant interest in academic and pharmaceutical communities for regulating cellular protein homeostasis. A series of MDM2 degraders were designed and synthesized, with WB214 identified as a potent anti-proliferative agent inducing the degradation of MDM2 and p53. Mechanistic studies revealed that WB214 functions as a molecular glue degrading MDM2, and MDM2 was discovered as a novel neo-substrate of the E3 ligase cereblon. Additionally, WB214 could potently degrade GSPT1, leading to the development of a selective degrader for GSPT1 over MDM2.
