M2 macrophage-conditioned medium inhibits intervertebral disc degeneration in a tumor necrosis factor-α-rich environment

Summary: This study reveals the therapeutic potential of M2 conditioned medium (M2CM) for intervertebral disc degeneration (IDD). M2CM from macrophages has anti-inflammatory and regenerative effects on nucleus pulposus cells (NPCs), promoting cell proliferation and extracellular matrix synthesis while inhibiting inflammation and apoptosis. These findings highlight the importance of M2CM in IDD treatment.

The degenerative impact of hyperglycemia on the structure and mechanics of developing murine intervertebral discs

Summary: Diabetes has been found to impact the biochemical composition, collagen fiber architecture, and mechanical behavior of intervertebral discs. Diabetic discs showed increased glycosaminoglycan and collagen content, as well as disorganized and unaligned collagen fibers at the boundary between the nucleus pulposus and annulus fibrosus. These changes made the diabetic discs stiffer and more resistant to deformation compared to wild-type discs. The findings suggest that diabetes may predispose individuals to degenerative disc disease later in life.

Measurement properties of grimace scales for pain assessment in nonhuman mammals: a systematic review

Summary: This systematic review aimed to assess the measurement properties of grimace scales for pain assessment in animals. The review found significant variability in the development and measurement properties of these scales across different species. The Mouse, Rat, Horse, and Feline Grimace Scales showed strong evidence, while the Rabbit, Lamb, Piglet, and Ferret Grimace Scales, as well as the Sheep Pain Facial Expression Scale, had moderate evidence. The Sheep Grimace Scale, EQUUS-FAP, and EQUUS-Donkey-FAP had low evidence for measurement properties. The most reported measurement property was construct validity, while reliability and other forms of validity were understudied.

The involvement of immune system in intervertebral disc herniation and degeneration

Summary: Intervertebral disc herniation and degeneration are closely related to low back pain, but the molecular pathogenesis is not fully understood. Inflammation plays a crucial role in this process, and the immune system may be involved. While the nucleus pulposus is an immune-privileged organ, the disruption of the physical barrier and the innervation and vascularization of the degenerated disc may lead to immune responses and pain generation.

Toward the Next Generation of Spine Bioreactors: Validation of an Ex Vivo Intervertebral Disc Organ Model and Customized Specimen Holder for Multiaxial Loading

Summary: A new generation of bioreactors with integrated six degrees of freedom (6 DOF) has been developed to mimic the natural load on intervertebral discs (IVD) more accurately. The study successfully developed and validated a specimen holder and corresponding ex vivo IVD organ model that met the requirements for multiaxial loading and long-term IVD culture. The study demonstrated that the developed holding system is mechanically reliable and biologically compatible for application in a new generation of multiaxial bioreactors.

Summary: Low back pain is a common cause of disability, with intervertebral disc degeneration being a major risk factor. Mouse models, due to their accessibility and ease of genetic manipulation, have been widely used in studying disc degeneration. Despite their small size, mouse lumbar discs have similarities to human discs in terms of geometry, structure, and mechanical properties. This article reviews current mouse models of intervertebral disc degeneration and identifies a set of critical output measurements for studying disease pathology and screening regenerative therapies, with a focus on pain phenotyping.
