Prevascularized Retrievable Hybrid Implant to Enhance Function of Subcutaneous Encapsulated Islets

Auvro R. Mridha et al.

Summary: This study successfully demonstrated the implantation of microencapsulated beta-cells in mice using a bioengineered hybrid device without the need for immunosuppressive drugs. Results showed that the device promoted vascularization and normalized blood glucose levels, providing a potential new approach for the treatment of type 1 diabetes in humans.

Enhanced Differentiation Capacity and Transplantation Efficacy of Insulin-Producing Cell Clusters from Human iPSCs Using Permeable Nanofibrous Microwell-Arrayed Membrane for Diabetes Treatment

In Kyong Shim et al.

Summary: This study developed a new platform using a biodegradable nanofibrous membrane to enhance the differentiation capacity and transplantation efficacy of insulin-producing cells (IPCs). The experimental results showed that the nanofibrous membrane promoted IPC cluster formation and improved insulin secretion. This platform has the potential to be used for in situ transplantation in diabetic patients.

Three-dimensional Vascularized β-cell Spheroid Tissue Derived From Human Induced Pluripotent Stem Cells for Subcutaneous Islet Transplantation in a Mouse Model of Type 1 Diabetes

Shohei Takaichi et al.

Summary: In this study, a vascularized beta-cell spheroid tissue was constructed using a layer-by-layer cell-coating technique and transplanted subcutaneously in a T1D model mouse. The results showed that this method could decrease blood glucose levels in diabetic mice, possibly due to host angiogenesis in the graft.

The Optimal Maturation of Subcutaneous Pouch Can Improve Pancreatic Islets Engraftment in Rat Model

Alzbeta Patikova et al.

Summary: The subcutaneous transplantation of pancreatic islets is less invasive yet equally effective as traditional portal vein transplantation. The procedure promotes vascularization and maintains normal blood glucose levels and body weight. Histological analysis indicates promising results with clusters of islet cells surrounded by rich vascular networks in the implanted scaffolds.

Pancreatic islet transplantation in type 1 diabetes: 20-year experience from a single-centre cohort in Canada

Braulio A. Marfil-Garza et al.

Summary: This study reports the long-term outcomes of islet transplantation in a single-center cohort, including patient survival, graft survival, insulin independence, glycemic control, and adverse events. The study found that the combined use of anakinra plus etanercept and the BETA-2 score were associated with improved outcomes.

Toward 3D-bioprinting of an endocrine pancreas: A building-block concept for bioartificial insulin-secreting tissue

Gabriel A. Salg et al.

Summary: Three-dimensional bioprinting of an endocrine pancreas is a promising future treatment for patients with insulin secretion deficiency. This study presents an end-to-end concept from the molecular to the macroscopic level, demonstrating the formation of viable and proliferative experimental models using a hybrid scaffold device and INS-I cells. Transcriptomics analysis reveals upregulation of cell proliferation and beta-cell-specific signaling as a result of pseudoislet formation and 3D-culture.

Portal Vein Thrombosis May Be More Strongly Associated With Islet Infusion Than Extreme Thrombocytosis After Total Pancreatectomy With Islet Autotransplantation

Alexander A. Boucher et al.

Summary: The study found that most patients developed extreme thrombocytosis after total pancreatectomy with islet autotransplantation (TPIAT), which was significantly associated with the infused islet volumes. The incidence of thromboembolic events was 12.2%, with portal vein thromboses occurring significantly earlier than peripheral thromboses.

Strategies for durable β cell replacement in type 1 diabetes

Todd M. Brusko et al.

Summary: Advancements in blood glucose monitoring and therapeutic insulin administration have improved the quality of life for individuals with type 1 diabetes, but still fall short of the metabolic control provided by native islets. Integrated advancements in islet cell replacement and immunomodulatory therapies are offering hope for the restoration of endogenous glucose regulation. Progress in stem cell biology and graft site design are providing innovative sources for cellular material and improved engraftment.
