Summary: c-Jun N-Terminal Kinases (JNKs), as members of the Mitogen-Activated Protein Kinase (MAPK) signaling pathway, play a crucial role in the pathogenesis of various diseases, making them attractive targets for therapeutic agents. Despite the discovery of many JNK inhibitors in the past decade, only a few have advanced to clinical trials due to the challenge of JNK-isoform selectivity.

Norisoboldine Attenuates Sepsis-Induced Acute Lung Injury by Modulating Macrophage Polarization via PKM2/HIF-1α/PGC-1α Pathway

Qi Chen et al.

Summary: Norisopoldine (NOR) effectively alleviated sepsis-induced acute lung injury (ALI) in mice by reducing inflammatory factors and promoting macrophage polarization from M1 to M2 type. NOR activated PKM2, inhibited its nuclear translocation, decreased HIF-1 alpha expression, and increased PGC-1 alpha and PPAR-gamma expression, leading to inhibition of glycolysis and promotion of oxidative phosphorylation in RAW264.7 cells. PKM2 inhibitors reversed the effects of NOR on the PKM2/HIF-1 alpha/PGC-1 alpha signaling pathway.

Dongyang Zhao et al.

Summary: Research on circN4bp1 in sepsis-induced ARDS patients and mice showed that overexpression of circN4bp1 is associated with poor prognosis, and its knockdown could improve lung injury by regulating macrophage activation. The study also revealed a circN4bp1/miR-138-5p network that affects macrophage polarization through EZH2 expression. Furthermore, m6A modification of circN4bp1 was found to be elevated in ARDS mice, indicating its potential as a therapeutic target or prognostic marker.
