The surface coating of iron oxide nanoparticles drives their intracellular trafficking and degradation in endolysosomes differently depending on the cell type

Summary: This study investigated the intracellular transit of two magnetic nanoparticles (MNPs) with different surface coatings in mouse cell lines. The results showed that the coating type influenced the intracellular trafficking rate and the nature of the endolysosomes in macrophages. The MNPs with a specific coating had a slower transit rate and resulted in endolysosomes with more lytic enzymes and catalytic proteins in macrophages. These MNPs also induced more autophagic vesicles and enhanced the expression of iron metabolism-related genes and proteins.

Cell Behavioral Changes after the Application of Magneto-Mechanical Activation to Normal and Cancer Cells

Summary: In vitro cell exposure to nanoparticles can help in the development of tools for detecting and treating human diseases. Recent studies have investigated the impact of magnetic field-actuated internalized magnetic nanoparticles on cancer cell behavior. The results showed that the viability rate of non-cancerous cells decreased mostly in cells with internalized nanoparticles and those treated with the pulsed magnetic field mode, while in cancer cells, the pulsed magnetic field mode along with internalized nanoparticles caused a significant decrease in viability.

Summary: Nanocomposite capsules containing magnetite nanoparticles have various biomedical applications. These capsules can be visualized using magnetic resonance imaging and optoacoustic imaging, and can be navigated and released remotely using magnetic fields. The magnetic moment of individual capsules, which is important for their behavior in magnetic fields, is determined by the amount of magnetite nanoparticles in the capsule. This study synthesized two types of submicron capsules with different amounts of magnetite nanoparticles and measured their magnetic moments using optical tweezers and vibrating-sample magnetometer measurements.

Magneto-mechanical destruction of cancer-associated fibroblasts using ultra-small iron oxide nanoparticles and low frequency rotating magnetic fields

Summary: By mechanically activating ultra-small superparamagnetic iron oxide nanoparticles with a rotating low-amplitude low-frequency magnetic field, cell death induction in pancreatic cancer-associated fibroblasts was maximized to 34%. The force generated by the nanoparticles inside lysosomes was calculated to be approximately 3 pN, leading to lysosome membrane permeabilization and cell death through a lysosomal pathway dependent on cathepsin-B activity.

Star-Shaped Magnetic-Plasmonic Au@Fe3O4 Nano-Heterostructures for Photothermal Therapy

Summary: Researchers synthesized a Au@Fe3O4 core@shell structure with a unique morphology and combined plasmonic and magnetic properties. Advanced electron microscopy characterization confirmed the excellent crystallinity and uniformity of the structure. The magnetite shell exhibited a higher saturation magnetization compared to previous Au@Fe3O4 structures, attributing to the star-like morphology and large shell thickness. Cancer cells loaded with Au@Fe3O4 nanostars displayed magneto mechanical stress under a low frequency external magnetic field. The homogeneous and thick magnetite shell shifted the plasmonic resonance of the Au core to 640 nm, enabling applications in photothermal therapy and optical imaging.
