Inflammageing and Cardiovascular System: Focus on Cardiokines and Cardiac-Specific Biomarkers

Marco Alfonso Perrone et al.

Summary: The term inflammageing was introduced in 2000 to describe the chronic inflammatory state of elderly individuals, which is characterized by elevated levels of inflammatory biomarkers, comorbidities, disability, frailty, and premature death. Inflammageing is linked to various cardiovascular diseases and early detection and counteraction of inflammageing can prevent cardiovascular disease and slow down age-related health decline. Early interventions require reliable and cost-effective biomarkers, such as cardiac-specific biomarkers, for diagnosis and further clinical studies are needed to assess their usefulness and cost/benefit ratio.

The Association of Age at Diagnosis of Hypertension with Cognitive Decline: the China Health and Retirement Longitudinal Study (CHARLS)

Linlin Ding et al.

Summary: This study investigated the association between age at diagnosis of hypertension and decline in cognitive performance. The study found that hypertension diagnosed in mid-life is associated with worse cognition compared to late life, and longer duration of hypertension is associated with worse memory. Blood pressure control is critical for the preservation of cognitive function.

Features of metabolic syndrome and inflammation independently affect left ventricular function early after first myocardial infarction

Jan Traub et al.

Summary: This study analyzed the relationships between BMI, other components of metabolic syndrome, and inflammatory markers with left ventricular geometry, function, and infarct size after ST-elevation MI. The results showed that BMI, HbA1c, and peak CRP were independent predictors of worse LVEF early after MI, while only peak CRP predicted worse LVEF at later stages.

Dysfunctional microglia and tau pathology in Alzheimer's disease

Summary: Extensive studies on humans and animals have shown that chronic immune system stimulation worsens neurodegeneration in diseases like Alzheimer's by activating microglia and promoting the production of harmful substances. Abnormalities in tau, A beta, and microglial activation are often observed in dementia patients and may contribute to cognitive impairment.

Hypertension and cognitive dysfunction: a narrative review

Eun-Jin Cheon

Summary: Cognitive dysfunction is a known complication of hypertension, with high blood pressure increasing the risk of cognitive decline and dementia. The most impacted cognitive functions in hypertensive individuals are executive functions and frontal lobe function, similar to vascular dementia. Cognitive disorders associated with hypertension include vascular dementia, Alzheimer's disease, and Lewy body dementia. The pathophysiology of cognitive dysfunction in hypertensive individuals includes brain atrophy, microinfarcts, microbleeds, neuronal loss, white matter lesions, network disruption, neurovascular unit damage, reduced cerebral blood flow, blood-brain barrier damage, enlarged perivascular damage, and proteinopathy. Antihypertensive drugs may help reduce the risk of cognitive decline and dementia.

Cognitive Impairment in Heart Failure: Landscape, Challenges, and Future Directions

Mengxi Yang et al.

Summary: Heart failure is a major healthcare problem globally and is often accompanied by cognitive impairment. The prevalence of cognitive impairment in heart failure patients is increasing and significantly affects their hospitalization and mortality rates. The current understanding of the mechanisms underlying cognitive impairment in heart failure is insufficient, and early diagnosis and intervention can improve prognosis. While clinical guidelines are starting to emphasize the importance of cognitive impairment, there is a lack of recommendations on how to manage it in heart failure patients.

Summary: There is an association between myocardial infarction and increased risk for vascular dementia, and both conditions have elevated inflammatory biomarkers linked to worse outcomes. However, the causative roles of specific cells in connecting cardiac injury to dementia development and progression are still not fully understood. This article provides an overview of the inflammatory cellular and molecular links between myocardial infarction and vascular dementia and discusses strategies to reduce inflammation after myocardial infarction to limit neurovascular injury.

Neuroinflammation inhibition by small-molecule targeting USP7 noncatalytic domain for neurodegenerative disease therapy

Xiao-Wen Zhang et al.

Summary: This study discovers a natural small molecule called eupalinolide B (EB) that inhibits USP7 to alleviate symptoms of neurodegenerative diseases. EB selectively modifies the Cys(576) site, inhibiting the activity of USP7 and causing the degradation of Keap1. This leads to the activation of anti-neuroinflammation genes in microglia. The effectiveness of this treatment method is validated in mouse models.

The study shows that TNFR1 plays a crucial role in suppressing prosurvival and proliferative signals in CD4+ T cells during heart failure, without altering their pathological activity.

Sustained Increase in Serum Glial Fibrillary Acidic Protein after First ST-Elevation Myocardial Infarction

Jan Traub et al.

Summary: The study found that serum levels of glial fibrillary acidic protein (GFAP) increased in the first 3 months after ST-elevation myocardial infarction (STEMI), and remained elevated without further increases after 6 and 12 months. In contrast, neurofilament light chain (NfL) levels remained unchanged throughout the one-year period. These findings support the idea of central nervous system involvement after myocardial infarction, with GFAP potentially serving as a peripheral biomarker for chronic glial damage.

Summary: The meninges contain a pool of monocytes and neutrophils supplied by adjacent skull and vertebral bone marrow, rather than by the blood. Therefore, myeloid cells from adjacent bone marrow niches are strategically placed to supply innate immune cells under both homeostatic and pathological conditions.
