Summary: The patient with progressive metastatic pancreatic cancer showed regression of visceral metastases after receiving infusion of genetically engineered autologous T cells targeting mutant KRAS G12D, with an overall partial response of 72% according to the Response Evaluation Criteria in Solid Tumors, version 1.1. The response was ongoing at 6 months, and the engineered T cells accounted for more than 2% of all circulating peripheral-blood T cells.

Summary: The number of Hematopoietic Cell Transplantations in Europe and collaborating countries has been on the rise, with a total of 48,512 HCT reported in 43,581 patients during 2019. The main indications were myeloid malignancies, lymphoid malignancies, and nonmalignant disorders. CAR-T cellular therapies also showed significant growth from 2017 to 2019. Over the past 30 years, there has been a substantial increase in the number of centers and countries performing HCT procedures, with more than 800,000 transplants reported overall.

Targeting a neoantigen derived from a common TP53 mutation

Summary: An antibody highly specific to the most common TP53 mutation has been identified and converted into an immunotherapeutic agent. Despite low complex density on the cancer cell surface, the bispecific antibody effectively activates T cells to lyse cancer cells expressing the neoantigen. This approach may be used to target cancers with mutations that are difficult to target conventionally.

A vaccine targeting mutant IDH1 in newly diagnosed glioma

Summary: Mutated isocitrate dehydrogenase 1 (IDH1) defines a molecularly distinct subtype of diffuse glioma. The most common IDH1 mutation in gliomas affects codon 132 and encodes IDH1(R132H), which harbours a shared clonal neoepitope that is presented on major histocompatibility complex (MHC) class II. An IDH1(R132H)-specific peptide vaccine (IDH1-vac) induces specific therapeutic T helper cell responses that are effective against IDH1(R132H)+ tumours.

This approach offers an efficient way to isolate antigen-specific TCRs for research and clinical use.
