In Situ Continuous Wave Electron Paramagnetic Resonance Investigation of the Amyloid Aggregation of Parkinson's Protein Alpha-Synuclein-the Second Spin-Label Position

Enrico Zurlo et al.

Summary: This study utilized spin-label EPR technique to investigate the impact of spin label position on aggregation kinetics and accessibility of local information on intermediates in αS aggregation. The results showed that rotation correlation times for the spin label in the oligomer cannot be determined accurately, and higher resolution EPR approaches, such as high-field EPR, are more promising at present stage.

In situ kinetic measurements of α-synuclein aggregation reveal large population of short-lived oligomers

Enrico Zurlo et al.

Summary: Understanding the mechanism of amyloid protein assembly is crucial for studying neurodegenerative diseases. EPR method can directly measure the amount of intermediates during the aggregation process without the need for separation, providing insights into the kinetics of intermediate species formation and conversion. The study on alpha-synuclein aggregation using EPR revealed a large population of short-lived primary oligomers formed directly from monomeric species.

Intracellular Protein-Lipid Interactions Studied by Rapid-Scan Electron Paramagnetic Resonance Spectroscopy

Theresa S. Braun et al.

Summary: Protein-membrane interactions are crucial for cellular processes, and the study of these interactions presents a challenge in modern biophysical chemistry. By employing rapid-scan electron paramagnetic resonance spectroscopy, the interaction between αS and negatively charged vesicles in vitro was studied, with reflections observed in cells as well.
