Anthracycline-induced cardiotoxicity and renin-angiotensin-aldosterone system-from molecular mechanisms to therapeutic applications

Pawel Sobczuk et al.

Summary: Millions of new cancer cases are diagnosed each year worldwide, and although advancements in cancer biology and therapies have reduced mortality rates, most patients still require chemotherapy, which can have high medical costs and adverse effects. Anthracyclines, commonly used in cancer treatment, are associated with a high risk of cardiotoxicity. The dysregulation of the renin-angiotensin-aldosterone system (RAAS), a major regulator of the cardiovascular system, may play a role in anthracycline-induced cardiotoxicity. Targeting this system with drugs has shown promise in the prevention and treatment of cardiac adverse events associated with anthracyclines.

Doxorubicin-induced apoptosis enhances monocyte infiltration and adverse cardiac remodeling in diabetic animals

Chandrakala Aluganti Narasimhulu et al.

Summary: This study aimed to investigate the cardiac toxicity induced by doxorubicin in diabetic animals and its underlying pathophysiological mechanisms. The results showed that compared with control and experimental groups, doxorubicin caused a significant increase in apoptosis, fibrosis, and structural alterations in diabetic animals.

Divergent Cardiac Effects of Angiotensin II and Isoproterenol Following Juvenile Exposure to Doxorubicin

Kevin Agostinucci et al.

Summary: This study aims to investigate whether pre-exposure to doxorubicin during adolescence would make the heart more susceptible to non-hypertensive pathologic stimuli. The results show that doxorubicin-induced latent cardiotoxicity enhances the cardiac response to hypertensive stimuli and is associated with worsened cardiac function and increased fibrosis.

Molecular mechanisms of doxorubicin-induced cardiotoxicity: novel roles of sirtuin 1-mediated signaling pathways

Summary: Doxorubicin is a commonly used chemotherapy drug, but its cardiotoxicity limits its clinical application. Recent studies have found that pre-treatment with dexrazoxane cannot fully improve the myocardial toxicity of doxorubicin, highlighting the need for more targeted cardioprotective strategies.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Combined Administration of Metformin and Atorvastatin Attenuates Diabetic Cardiomyopathy by Inhibiting Inflammation, Apoptosis, and Oxidative Stress in Type 2 Diabetic Mice

Weikun Jia et al.

Summary: The combination therapy of metformin and atorvastatin showed superior protective effects against diabetic cardiomyopathy by inhibiting oxidative stress and inflammation, as well as reducing apoptosis of cardiomyocytes. This effect may be related to the activation of AMPK/SIRT1 signaling pathway.

Serial Cardiovascular Magnetic Resonance Strain Measurements to Identify Cardiotoxicity in Breast Cancer Comparison With Echocardiography

Christian P. Houbois et al.

Summary: In early stage breast cancer patients, changes observed in CMR and 2DE strain, left ventricular ejection fraction, and LVESVi can be used as prognostic indicators for subsequent CTRCD. Two-dimensional echocardiography strain measurement (2DE-GLS) provided the highest increase in discriminatory value over baseline clinical risk factors for subsequent CTRCD.

Summary: Cardiotoxicity in late phase after anthracycline drug administration remains unclear. Among patients receiving anthracycline treatment, some may develop cancer therapeutics-related cardiac dysfunction (CTRCD). Cardiac computed tomography (CCT) may be a comparable tool to echocardiography for evaluating CTRCD.
