Small-diameter artery decellularization: Effects of anionic detergent concentration and treatment duration on porcine internal thoracic arteries

Summary: Engineered replacement materials have great potential in vascular applications with a focus on optimizing decellularization processes for xenogeneic vascular tissue sources. Different anionic detergent concentrations were investigated for their ability to effectively remove cell proteins while leaving DNA content mostly intact. Longer treatment times improved decellularization by significantly reducing DNA content, but also led to reduced laminin content and altered mechanical properties of the vessels.

Summary: The current research on small-diameter tissue-engineered vascular grafts (SD-TEVGs) focuses on improving design to address issues like mechanical failure, thrombogenicity, and intimal hyperplasia that have limited success so far. Synthetic polymer-based SD-TEVGs have shown limited success in large-animal models, while natural polymer and cell-secreted ECM-based SD-TEVGs are still facing challenges in terms of strength and thrombogenicity. Further exploration of allogeneic native ECM-based SD-TEVGs, in vitro-developed hybrid SD-TEVGs with allogeneic banked human cells or isolated autologous stem cells, and in-body tissue architecture (IBTA)-based SD-TEVGs appear promising for the future due to their dimension, mechanical strength, biocompatibility, and availability.
