NOTCH Signaling in Aortic Valve Development and Calcific Aortic Valve Disease

Yidong Wang et al.

Summary: NOTCH signaling regulates multiple biological processes involved in tissue morphogenesis and homeostasis. Mutations in NOTCH1 can cause congenital bicuspid aortic valve and calcific aortic valve disease, with genetic variants affecting other genes in the pathway also contributing to these diseases. The role of NOTCH signaling in the postnatal homeostasis of the aortic valve is crucial, as animal studies have provided detailed insights into the developmental etiology of congenital aortic valve defects.

Multiplexed imaging mass spectrometry of the extracellular matrix using serial enzyme digests from formalin-fixed paraffin-embedded tissue sections

Cassandra L. Clift et al.

Summary: This study presents a multiplexed imaging mass spectrometry method to spatially localize and selectively access the extracellular matrix on formalin-fixed paraffin-embedded tissue sections. A novel serial enzyme strategy has been developed to define the extracellular matrix, including PTMs, from a single tissue section for MALDI-IMS applications to overcome the challenge of capturing complex ECM peptides.

Summary: Calcific Aortic Valve Disease (CAVD) is the most common valvular heart disease in the ageing population, which is strongly correlated with active and degenerative processes. The progression of the disease is closely related to the cellular and matrix alterations within the aortic valve.

Multi-Omics Approaches to Define Calcific Aortic Valve Disease Pathogenesis

Mark C. Blaser et al.

Summary: Calcific aortic valve disease is expected to almost triple in prevalence over the next 3 decades and currently lacks effective pharmacotherapy to halt its progression. Invasive aortic valve replacement remains the only treatment option, highlighting a substantial gap in care due to limited understanding of disease mechanisms and complex valvular microenvironment. Multiomics approaches, single-cell resolution studies, and systems biology strategies hold promise in shedding new light on valvular disease pathogenesis and identifying potential drug targets.

Spatial N-glycomics of the human aortic valve in development and pediatric endstage congenital aortic valve stenosis

Peggi M. Angel et al.

Summary: This study investigated the spatial localization of N-glycan structures in pediatric congenital aortic valve stenosis. The N-glycome was found to be localized in different parts of the normal aortic valve and hypertrophic commissures in AVI. Sialic acid N-glycans were the most abundant group in all valve types, with common elevation in AS independent of age.

Summary: The study developed collagenous tubes that can grow with children and successfully implanted them in lambs as pulmonary artery replacements. The tri-tube valved conduits showed better growth and functionality compared to bioprosthetic conduits in two sequential cohorts, with reduced calcification and improved hemodynamic function observed in the lamb models.

Collagen fiber regulation in human pediatric aortic valve development and disease

Cassandra L. Clift et al.

Summary: This study investigates collagen fiber production and regulation in human AV development and pediatric end-stage CAVS by combining histological studies with high resolution accurate mass (HRAM) collagen-targeting proteomics. Specific collagen peptides modified by hydroxylated prolines (HYP) were found to be significantly regulated across patient categories. The study also identified non-collagen type ECM proteins which have direct interactions in collagen synthesis, regulation, or modification, suggesting potential therapeutic avenues for inhibiting valvular degradation and engineered options for valve replacement in pCAVS.

Marialucia Gallorini et al.

Summary: Collagen, as the most abundant protein in the extracellular matrix, is widely used in various industries due to its biocompatible properties. Recent advancements in understanding collagen functions have revealed additional roles beyond scaffolding and mechanotransduction, leading to exploration of small molecules and peptides for therapeutic purposes. However, challenges in targeting collagen interactions remain due to the presence of multiple isoforms and lack of specific 3D crystal structures.

Summary: This study found that aortic valve calcification is associated with disruption in extracellular matrix and metabolic pathways, highlighting a strong connection between metabolic markers and ECM remodeling. Additionally, FNDC1 and MXRA5 were identified as novel ECM biomarkers in calcified valves, potentially serving as targets in the development and progression of aortic stenosis.
