A gain-of-function mutation in the ITPR1 gating domain causes male infertility in mice

Bo Sun et al.

Summary: ITPR1 is a critical intracellular calcium release channel that is involved in numerous cellular processes. While mutations in ITPR1 have been primarily associated with movement disorders, our study reveals the pathological consequences of ITPR1 gain of function (GOF) mutations, which enhance ITPR1 activity and increase the risk of testicular dysfunction.

Summary: This study presents cryo-EM structures of human type-3 IP3 receptors, revealing the molecular mechanism of receptor activation. The structures demonstrate how IP3-induced conformational changes prime the receptor for activation by Ca2+, and how ATP modulates the activity.

Omnitemporal choreographies of all five STIM/Orai and IP3Rs underlie the complexity of mammalian Ca2+ signaling

Scott M. Emrich et al.

Summary: The study reveals the non-redundant functions of STIM1/2 and Orai1/2/3 proteins in ensuring the graded diversity of mammalian Ca2+ signaling. Additionally, it shows that the unactivated STIM1/2 inhibit IP3R-evoked Ca2+ release, with the inhibition gradually relieved as agonist intensity and STIM1/2 activation increase.

KRAP tethers IP3 receptors to actin and licenses them to evoke cytosolic Ca2+ signals

Nagendra Babu Thillaiappan et al.

Summary: The study demonstrates that the KRas-induced KRAP protein tethers a subset of IP3 receptors to actin, enabling them to evoke calcium signals. Loss of KRAP results in the disappearance of Ca2+ puffs and a global response to increased cytosolic Ca2+ concentration from more intense stimulation.
