High-Fidelity Drug-Induced Liver Injury Screen Using Human Pluripotent Stem Cell-Derived Organoids

Tadahiro Shinozawa et al.

Summary: This study established a screening model based on human liver organoids (HLO) for analyzing drug-induced liver injury (DILI) pathology, providing unique advantages at the organoid resolution. Through this model, a multi-readout organoid analysis was successfully developed to measure viability, cholestatic and/or mitochondrial toxicity, showing high predictive values for marketed drugs.

Sequential Activation of Guide RNAs to Enable Successive CRISPR-Cas9 Activities

Ryan Clarke et al.

Summary: The system presented in this study demonstrates a new approach to executing multiple genetic manipulations as a preprogrammed cascade of events, without the need for engineering cell-type-specific promoters or gene regulatory sequences. By leveraging the programmability of S. pyogenes Cas9 and arranging modules based on sgRNA, it allows for sequential genetic manipulations in a programmable manner that interfaces with mammalian cell biology at multiple levels.

Hepatocellular carcinoma

Josep M. Llovet et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), poses a significant global health challenge with rising incidence projected to exceed 1 million cases by 2025. Infection by hepatitis B and C viruses remains a major risk factor for HCC, while non-alcoholic steatohepatitis is emerging as a more common risk factor in Western countries. Advances in systemic therapies for HCC, including immunotherapies and targeted therapies, are expected to revolutionize the management of this disease.

A 3D microfluidic liver model for high throughput compound toxicity screening in the OrganoPlate®

Kristin M. Bircsak et al.

Summary: This study presents the development, automation, and validation of a 3D, microfluidic liver-on-a-chip system for high-throughput hepatotoxicity screening. The model consists of iPSC-derived hepatocyte aggregates co-cultured with endothelial cells and macrophages in a microchannel platform. With automation protocols and robust statistical analysis, the model demonstrated stable cell viability and functionality over time, making it a promising tool for hepatotoxicity screening and compound prioritization.

Synthetic liver fibrotic niche extracts achieve in vitro hepatoblasts phenotype enhancement and expansion

Yuying Zhang et al.

Summary: The challenge of synthesizing 'cellular niche-mimics' in vitro with satisfactory reproducibility and fidelity to recreate natural niche components for stem cell cultivation remains. Inspired by liver fibrosis-induced amplification of hepatic progenitor cells, researchers optimized fibrotic niches in vitro and harvested bioactive ingredients as niche extracts. These extracts outperformed Matrigel in maintaining human embryonic stem cells and replicating pathological angiogenesis, providing innovative strategies for discovering optimal cellular niche compositions.
