In-situ high-resolution 3D imaging combined with proteomics and metabolomics reveals enlargement of subcellular architecture and enhancement of photosynthesis pathways in nuclear-irradiated Chlorella pyrenoidosa

Summary: Trebouxiophyceae is a diverse class of green algae found in terrestrial and aquatic environments. Among them, the genus Trebouxia is widely distributed in lichens worldwide. However, the current species descriptions do not fully represent the overall diversity of the genus. By studying the ultrastructure of pyrenoids and the morphology of chloroplasts, it is possible to differentiate and characterize species-level lineages effectively.

Our analysis using electron microscopy revealed differentiated plasma membrane structures called nubbins in lichens, which play a role in cell polarity determination and inter-hyphal communication. Additionally, we found that substances in algal growth medium can induce fungal differentiation.

Summary: Near-infrared spectroscopy (NIRS) was used to characterize the metabolomic fingerprints of lichens and cultured microalgae. The study found that different water structures, lipids, and sugars contribute to the composition of lichens and microalgae. Lichen species with higher desiccation tolerance have higher levels of bound water, while fresh microalgae are mainly composed of free water. Aquaphotomics reveals the involvement of strongly hydrogen bonded water structures, polyols/glucides, and unsaturated/saturated fatty acids in the dehydration process.
