Pretomanid for tuberculosis: a systematic review

Tinne Gils et al.

Summary: Pretomanid shows potential efficacy in the treatment of tuberculosis, especially in rifampicin-resistant and highly resistant tuberculosis. However, serious hepatotoxic adverse events and drug-related toxicities were reported, indicating the need for further evaluation and comparison with existing drugs.

Human ancient DNA analyses reveal the high burden of tuberculosis in Europeans over the last 2,000 years

Gaspard Kerner et al.

Summary: This study revealed the history of human exposure to tuberculosis in Europe and found that the TYK2 P1104A variant originated from common ancestors of West Eurasians around 30,000 years ago. The frequency of the variant fluctuated significantly over the past 10,000 years, with a dramatic decrease after the Bronze Age. The drop in frequency was attributed to strong negative selection starting around 2,000 years ago, resulting in a 20% relative fitness reduction in homozygotes, indicating a heavy burden of tuberculosis on European health over the last two millennia.

A high-resolution HLA reference panel capturing global population diversity enables multi-ancestry fine-mapping in HIV host response

Yang Luo et al.

Summary: By constructing a high-resolution reference panel based on whole-genome sequencing data, accurate imputation of HLA alleles across diverse populations and fine-mapping of HLA association signals for HIV-1 host response were achieved. The study demonstrated accurate imputation of HLA alleles in various global populations, and identified a novel association at position 156 in the HLA-B gene.

First description of the katG gene deletion in a Mycobacterium tuberculosis clinical isolate and its impact on the mycobacterial fitness

Flavio De Maio et al.

Summary: This study identified a clinical Mtb strain with a complete deletion of the katG gene, conferring high-level resistance to INH, while not impacting the strain's fitness and pathogenic potential.

Adjunctive host-directed therapies for pulmonary tuberculosis: a prospective, open-label, phase 2, randomised controlled trial

Robert S. Wallis et al.

Summary: This study conducted a phase 2 clinical trial in South Africa to evaluate the safety and preliminary efficacy of four host-directed therapies for tuberculosis. The results showed that CC-11050 and everolimus as adjunctive therapies have the potential benefits for tuberculosis patients, increasing the recovery of FEV1.

Efficacy of bedaquiline in the treatment of drug-resistant tuberculosis: a systematic review and meta-analysis

Ming-Gui Wang et al.

Summary: The study demonstrated that bedaquiline treatment is associated with higher culture conversion rates and lower mortality risks among patients with drug-resistant tuberculosis when compared to those who do not receive bedaquiline. However, there was no significant improvement in treatment success rates.
