Structure-Activity Relationships for 5′′ Modifications of 4,5-Aminoglycoside Antibiotics

Summary: Modification at the 5''-position of 4,5-disubstituted aminoglycoside antibiotics can circumvent inactivation by aminoglycoside modifying enzymes, but its impact on drug activity and target selectivity is unpredictable. In this study, various modifications at the 5''-position were investigated and it was found that a hydrogen bonding group is necessary for maintaining antibacterial activity in certain compounds, while the installation of a formamido or ureido group can preserve both activity and selectivity.
