Characterization of a Novel Antimicrobial Peptide Isolated from Moringa oleifera Seed Protein Hydrolysates and Its Membrane Damaging Effects on Staphylococcus aureus

Summary: The present study identified and characterized a novel antimicrobial peptide, MOp2, from Moringa oleifera seed protein. MOp2 showed potential antimicrobial effects on Staphylococcus aureus, with negligible hemolytic activity and maintained antimicrobial activity under different conditions. Additionally, MOp2 caused membrane damage to S. aureus cells and interacted with key enzymes in bacteria to inhibit growth.

Rational design of hyperstable antibacterial peptides for food preservation

Summary: This study describes the design of peptides with thermostability, pH stability, and antibacterial activity against foodborne pathogens. Through classical structure-function analysis and mutational studies, a set of peptides were developed and demonstrated robust antimicrobial activity even under high temperatures and wide pH ranges. The mode of action of these hyperstable peptides was investigated through membrane permeability assays, electron microscopy, and molecular dynamics simulations, revealing that they exert antibacterial action through both membrane destabilization and inhibition of intracellular trypsin. Toxicity studies and food preservation assays confirmed the safety and efficacy of these designed peptides for food preservation. Insights from this work may be valuable for future development of antimicrobials.
