Microalgal metabolic engineering strategies for the production of fuels and chemicals

Nam Kyu Kang et al.

Summary: Microalgae are promising sustainable resources that can convert CO2 into biofuels and chemicals directly, but their industrial production and economic feasibility are still limited. Metabolic engineering approaches have been used to enhance the productivity of microalgal bioproducts, with impressive advances in the last decade. Challenges remain in achieving effective production of chemicals and fuels via microalgal metabolic engineering.

Salt induced oxidative stress alters physiological, biochemical and metabolomic responses of green microalga Chlamydomonas reinhardtii

Soufiane Fal et al.

Summary: This study investigated the physiological, biochemical, and metabolomic changes in the microalga Chlamydomonas reinhardtii under salt stress. The results showed that salt stress resulted in decreased growth rate and chlorophyll levels, as well as increased ROS generation and lipid peroxidation. Moreover, the microalga exhibited increased carbohydrate and lipid accumulation, along with higher levels of carotenoids, polyphenols, and osmoprotectant molecules. The study also revealed changes in enzyme activities and the accumulation of specific molecules related to membrane remodeling and stress resistance.

Fatty acid export (FAX) proteins contribute to oil production in the green microalga Chlamydomonas reinhardtii

Janick Peter et al.

Summary: In this study, the researchers found that Cr-FAX1 and Cr-FAX5 are key proteins involved in TAG production in Chlamydomonas, located in the chloroplast and ER membranes respectively. Decreasing the expression of these proteins led to a significant reduction in TAG content. Additionally, overexpression of Cr-FAX1 in the chloroplast doubled the TAG content.

Enhanced accumulation of oil through co-expression of fatty acid and ABC transporters in Chlamydomonas under standard growth conditions

Ru Chen et al.

Summary: This study demonstrates that co-overexpression of chloroplast and endoplasmic reticulum-localized fatty acid transporters in Chlamydomonas enhances lipid accumulation and metabolism, particularly the remobilization of polyunsaturated fatty acids. The findings suggest a potential strategy for increasing the production of energy-rich and value-added compounds in microalgae.

ROS Induce β-Carotene Biosynthesis Caused by Changes of Photosynthesis Efficiency and Energy Metabolism in Dunaliella salina Under Stress Conditions

Yimei Xi et al.

Summary: This study investigated the regulatory mechanism of beta-carotene accumulation in Dunaliella salina under stress conditions, revealing a positive correlation between cellular ROS level and beta-carotene content, and a negative correlation between F-v/F-m of PSII and beta-carotene content. Transcriptional analysis showed that ROS induce beta-carotene accumulation by modulating genes involved in photosynthesis and beta-carotene biosynthesis.

The disassembly of lipid droplets in Chlamydomonas

Yonghua Li-Beisson et al.

Summary: Lipid droplets are specialized organelles in eukaryotic cells, consisting of a triacylglycerol core surrounded by a monolayer of membrane lipids. Their functions range from energy storage to signal transduction, and their turnover is tightly coordinated with cellular metabolic needs in a fluctuating environment. In Chlamydomonas reinhardtii, key components of lipid droplet turnover have been identified, highlighting the physiological significance of this process in algal oil research.
