Serine pseudoproteases in physiology and disease

Nina Zupanic et al.

Summary: Serine proteases play a crucial role in physiology and pathology. The discovery of nonenzymatic functions in pseudo serine proteases (SPHs) has expanded our understanding of the role of these proteins. SPHs have been found to be important in reproduction, embryonic development, immune response, host defense, and hemostasis. Investigating the interaction between SPs and SPHs can uncover new physiological functions and aid in the development of innovative medical approaches.

The Piroplasmida Babesia, Cytauxzoon, and Theileria in farm and companion animals: species compilation, molecular phylogeny, and evolutionary insights

Leonhard Schnittger et al.

Summary: The order Piroplasmida, consisting of Babesia, Cytauxzoon and Theileria, are tick-transmitted hemoprotozoans causing serious impact in domestic animals and humans. International research is conducted to identify, classify and develop treatments for these parasites. This review presents a compilation of piroplasmid species, isolates and species complexes that infect domestic mammals and demonstrates that there may be more piroplasmid parasites than vertebrate host species. The study also reveals insights into the evolutionary patterns and host-parasite relationships of these parasites.

Unraveling the Complexity of the Rhomboid Serine Protease 4 Family of Babesia bovis Using Bioinformatics and Experimental Studies

Romina Gallenti et al.

Summary: This study investigated the ROM4 coding genes of the tick-transmitted protozoon Babesia bovis. The expression of ROM4 genes in different stages and the genetic polymorphisms among geographic isolates were analyzed. The findings suggest that ROM4 likely plays an important role in host-pathogen interactions and may serve as a potential target for control strategies against this pathogen.

Babesia, Theileria, Plasmodium and Hemoglobin

Daniel Sojka et al.

Summary: This paper provides an updated comparison of the feeding mechanisms during the intraerythrocytic stage of Plasmodium spp. and Babesia/Theileria spp., two distantly related groups of parasitic Apicomplexa. It highlights fundamental differences in metabolic pathways and protein degradation, which have important implications for managing and intervening in Babesia/Theileria infections.

The repertoire of serine rhomboid proteases of piroplasmids of importance to animal and human health

Romina Gallenti et al.

Summary: This study classified and analyzed rhomboid proteins encoded in the genomes of different piroplasmid species, identifying 44 incorrectly classified or misannotated rhomboids. The distinguishing feature of Babesia s.s. from other piroplasmid lineages is the increased number of ROM4 paralogs. Piroplasmid ROM6 is the only rhomboid that is ubiquitous in other organisms.

Interaction between transforming Theileria parasites and their host bovine leukocytes

Shahin Tajeri et al.

Summary: Theileria are tick-transmitted parasites that cause fatal leuko-proliferative diseases in cattle, but their transformation of leukocytes is reversible with anti-theilerial drug treatment. Theileria-transformed leukocytes disseminate in infected cattle causing cancer-like disease. Cytokines, noncoding RNAs, and oncometabolites may contribute to the transformed phenotype and disease pathology.

Derlin rhomboid pseudoproteases employ substrate engagement and lipid distortion to enable the retrotranslocation of ERAD membrane substrates

Anahita Nejatfard et al.

Summary: A recent study identified a new protein, Dfm1, that plays a key role in retrotranslocation process, with conserved residues critical for this function. Additionally, it was found that Dfm1 possesses lipid thinning function to facilitate the removal of ER membrane substrates, a feature also conserved in its human homolog, Derlin-1.

Plasmepsin-like Aspartyl Proteases in Babesia

Pavla Snebergerova et al.

Summary: Apicomplexan genomes encode multiple pepsin-family aspartyl proteases that have diversified to adapt to different strategies of host infection and transmission. Expressional profiling of Babesia microti APs and phylogenetic analysis of APs from selected Babesia and Theileria species indicate their potential roles and druggable enzymatic targets for specific therapy for babesiosis.

In Silico Survey and Characterization of Babesia microti Functional and Non-Functional Proteases

Monica Florin-Christensen et al.

Summary: Human babesiosis caused by Babesia microti is an expanding tick-borne zoonotic disease that may cause severe symptoms and death, with a lack of therapeutic alternatives due to drug resistance. The study identified the full repertoire of proteases in the B. microti genome and revealed differences in protease repertoire among different Babesia parasites, providing potential targets for novel therapeutic interventions.

The FtsHi Enzymes of Arabidopsis thaliana: Pseudo-Proteases with an Important Function

Laxmi S. Mishra et al.

Summary: FtsH metalloproteases play a vital role in the maintenance and proteolysis of membrane proteins in various organisms, with some pseudo-proteases being inactive due to mutations or missing motifs. In Arabidopsis thaliana, these proteases are involved in plant growth and development, as well as oxidative stress management.
