Tracking changes in national BCG vaccination policies and practices using the BCG World Atlas

Samantha Lancione et al.

Summary: The BCG vaccine has varying effectiveness and policies depending on the country, and the third BCG World Atlas provides information on BCG practices in 194 countries. Many countries have shifted to targeted vaccination for high-risk groups and have seen a decrease in TB incidence. Despite a manufacturing interruption in 2015, the Danish strain remains the most commonly used BCG strain globally. Tracking and understanding changes in national BCG practices and their impact on TB burden is crucial for future immunization guidelines.

Ultra-low Dose Aerosol Infection of Mice with Mycobacterium tuberculosis More Closely Models Human Tuberculosis

Courtney R. Plumlee et al.

Summary: The study successfully developed a tuberculosis model in mice that more closely resembles human disease by infecting them with an ultra-low dose of bacteria, resulting in highly heterogeneous bacterial burdens and granulomas similar to those in humans. By identifying blood RNA signatures, it was possible to predict infection outcomes in mice and infer the risk of progression to active tuberculosis in humans.

The safety of BCG revaccination: A systematic review

Samantha Bannister et al.

Summary: The review found that revaccination with BCG vaccine carries minimal risk for immunocompetent individuals, with mainly mild local and systemic reactions. Future studies should focus on reporting adverse event data for novel applications of BCG vaccine.

High-dose Mycobacterium tuberculosis aerosol challenge cannot overcome BCG-induced protection in Chinese origin cynomolgus macaques; implications of natural resistance for vaccine evaluation

Laura Sibley et al.

Summary: This study demonstrated that the BCG vaccine is effective in controlling high dose aerosol Mycobacterium tuberculosis challenge in cynomolgus macaques of Chinese origin (CCM). The study also revealed that low levels of IFN-γ induced by the BCG vaccination in CCM, together with other factors, contribute to the natural ability of CCM to control infection.

TB and SIV Coinfection; a Model for Evaluating Vaccine Strategies against TB Reactivation in Asian Origin Cynomolgus Macaques: A Pilot Study Using BCG Vaccination

Andrew D. White et al.

Summary: This pilot study demonstrated the utility of a cynomolgus macaque model for evaluating vaccines designed to prevent reactivation of TB through coinfection with SIV. Results showed that BCG vaccination improved control of TB-induced disease and reduced viral load in coinfected animals.
