Notable sphingolipids like ceramide and sphingosine-1 phosphate have contrasting effects on cell survival and chemotherapeutic resistance, with ceramide inducing death and lethal mitophagy, and sphingosine-1 phosphate promoting survival. These lipids are characterized by a long-chain sphingosine backbone linked to a fatty acyl chain with variable length, and alterations in ceramide synthases and sphingolipid regulation of mitophagy have important implications for cancer therapy.
