Summary: African swine fever virus (ASFV) causes significant morbidity and mortality in pigs worldwide. This study developed a live attenuated ASFV mutant by deleting two interferon inhibitors and found that the mutant was safe and highly resistant to genetic modification. High doses of the mutant provided sterile immunity and complete protection in a virulent challenge. Two doses were superior to one dose and provided additional protection. This research contributes to the development of a potential vaccine option against ASF.

Classification and characterization of multigene family proteins of African swine fever viruses

Summary: This study classified MGF proteins into 31 groups based on sequence homology and network clustering, revealing that MGF groups of the same family were most similar to each other and had conserved sequence motifs. The genetic diversity of MGF groups varied widely mainly due to indels, while MGF proteins of the same family tended to have similar structure, location, and function. Reconstruction of the ancestral states of MGF groups along the ASFV phylogeny showed that most MGF groups experienced copy number variations or gain-or-loss changes, with a tendency towards loss in MGF proteins during evolution.
