Summary: We found that SARS-CoV-2 infection in Syrian hamsters leads to the activation of the hypoxia-inducible factor (HIF-1 alpha) transcriptional pathway, possibly due to hypoxia or accumulation of mitochondrial reactive oxygen species (ROS) in the lungs. This activation results in increased expression of HIF-1 alpha target proteins and a rise in glycolysis, while mitochondrial respiration decreases. Proteomic analysis also revealed specific deficits in mitochondrial ATP synthase and ATP/ADP translocase. Activation of HIF-1 alpha in inflammatory macrophages may drive inflammation, proinflammatory cytokine production, complement activation, and oxidative stress in infected lungs. These findings suggest a central role for HIF-1 alpha in the metabolic reprogramming, inflammation, and bioenergetic dysfunction associated with SARS-CoV-2 infection.

Microglia ferroptosis is regulated by SEC24B and contributes to neurodegeneration

Summary: Iron dysregulation is associated with multiple neurodegenerative diseases, including Parkinson's disease. In this study, it is found that iron-loaded microglia are responsive to iron and susceptible to ferroptosis, a form of cell death that is dependent on iron. Iron overload also induces a shift in the microglial transcriptional state that is similar to the transcriptomic signature found in postmortem brain microglia of Parkinson's disease. The removal of microglia delays iron-induced neurotoxicity, suggesting the contribution of microglial response to neurodegeneration.

Empagliflozin Improves the MicroRNA Signature of Endothelial Dysfunction in Patients with Heart Failure with Preserved Ejection Fraction and Diabetes

Summary: Endothelial dysfunction is a key mechanism in heart failure with preserved ejection fraction (HFpEF), diabetes mellitus (DM), and frailty, but there is a lack of reliable biomarkers to monitor this dysfunction. This study evaluated the expression of circulating microRNAs involved in endothelial function regulation in HFpEF and DM patients treated with empagliflozin, metformin, or insulin. The results showed that specific miRNAs were significantly regulated in HFpEF patients, and two of them (miR-21 and miR-92) were significantly reduced after empagliflozin treatment.

Exosomal miR-145 and miR-885 Regulate Thrombosis in COVID-19

Summary: We hypothesized that exosomal microRNAs may play a role in the development of thromboembolic complications in COVID-19. We isolated circulating exosomes from COVID-19 patients and categorized them based on D-dimer levels. We found that exosomal miR-145 and miR-885 were significantly correlated with D-dimer levels. Furthermore, human endothelial cells treated with serum from COVID-19 patients showed reduced release of miR-145 and miR-885, increased apoptosis, and impaired angiogenetic properties compared to cells treated with non-COVID-19 serum.
