Recognition of ATT Triplex and DNA:RNA Hybrid Structures by Benzothiazole Ligands

Summary: Interactions between a series of benzothiazole ligands and nucleic acid structures were screened using competition dialysis. Two compounds demonstrated high affinities against 13 nucleic acid structures, while another compound showed high selectivity among the studied compounds. Additionally, some compounds exhibited specific effects on certain nucleic acid structures.

Benzoselenazolium-based hemicyanine dye for G-Quadruplex detection

Summary: In this study, a benzoselenazolium-based hemicyanine dye was developed for the selective detection of G-quadruplexes. The dye exhibited red fluorescence in the presence of G-quadruplexes and could be used to detect nucleolar and mitochondrial G-quadruplexes.

Styryl dyes with N-Methylpiperazine and N-Phenylpiperazine Functionality: AT-DNA and G-quadruplex binding ligands and theranostic agents

Summary: New monomethine, unsymmetrical styryl dyes consisting of benzothiazole and N-methylpiperazine or N-phenylpiperazine scaffolds were synthesized and studied for their binding affinities for different ds-polynucleotides and G-quadruplex. The substitution of piperazine unit strongly influenced their binding modes, affinities, spectroscopic responses, and antiproliferative activities. N-methylpiperazine-substituted compounds showed a preference for AT-DNA polynucleotides and demonstrated AT-minor groove binding, while N-phenylpiperazine substituted compounds showed stronger stabilization of G-quadruplex. Fluorimetric titrations suggested possible mixed stoichiometries with G-quadruplex. Compounds with N-phenylpiperazine fragments exhibited micromolar and submicromolar antiproliferative activity, particularly against leukemia and lymphoma.

A Selenium-Substituted Heptamethine Cyanine Photosensitizer for Near-Infrared Photodynamic Therapy

Summary: In this study, we successfully developed a near-infrared photosensitizer with a long-lived excited triplet state, which exhibits significantly high singlet oxygen quantum yield. Good photodynamic therapy effect was demonstrated in living cell experiments. The selenium-substituted photosensitizer has low dark cytotoxicity and good chemical stability.

Formation of triplex rA/dA-containing nucleic acid helices induced by new thiazole orange analogues and their biological evaluation. H-aggregate formation conditioned by rA sequence

Summary: A series of dicationic monomethine cyanine dyes were synthesized with structural alterations to examine their interactions with DNA and RNA. The chlorinated cyanine dye showed strong stability in triplex helices and tendency for H-aggregate formation. These dyes exerted a significant effect on tumor cell viability, especially in HeLa and A549 cell lines. Fluorophores 2 and 3 preferentially accumulated in mitochondria and showed promising therapeutic potential.

An Insight into Symmetrical Cyanine Dyes as Promising Selective Antiproliferative Agents in Caco-2 Colorectal Cancer Cells

Summary: This study synthesized a series of symmetric cyanine dyes and evaluated their potential as anticancer agents. The dyes demonstrated high antiproliferative effects on several cancer cell lines in the absence of light, with one dye derived from benzoxazole showing the most promise. It had a high selectivity index and could arrest the cell cycle.

Summary: A series of asymmetric monomeric monomethine cyanine dyes exhibit nucleic acids staining ability with ratiometric red shift of their longer wavelength absorption maxima. The dyes show negligible fluorescence in water solutions but become strongly fluorescent after contacting with nucleic acids, especially with RNA. Molecular docking studies reveal that 1a-1c are groove binders while 2b-2c interact with DNA through partial intercalation mode. The nature of electronic transitions and their manifestation in the electronic spectra are revealed by DFT and TDDFT calculations.
