Differentially targeted seeding reveals unique pathological alpha-synuclein propagation patterns

Shady Rahayel et al.

Summary: The study investigates the long-term spatiotemporal distribution of Lewy-related pathology in mice injected with alpha-synuclein preformed fibrils and recreates these patterns using a computational model. The findings suggest that both regional connectivity and Snca gene expression play a significant role in the propagation of pathologic alpha-synuclein.

Phosphorylation of endogenous α-synuclein induced by extracellular seeds initiates at the pre-synaptic region and spreads to the cell body

Shiori Awa et al.

Summary: This study investigated the early phase of phosphorylated alpha-synuclein accumulation by injecting alpha-synuclein fibrils into the brains of wild-type mice. The results showed that phosphorylated alpha-synuclein appeared within a few days after injection, initially in presynaptic regions and subsequently extending its localization to axons and cell bodies.

Similar neuronal imprint and no cross-seeded fibrils in α-synuclein aggregates from MSA and Parkinson's disease

Florent Laferriere et al.

Summary: The principal component of Lewy bodies and glial cytoplasmic inclusions is aggregated alpha-synuclein, and the inclusion bodies may cause cell death through the sequestration of partner proteins and organelles. Furthermore, the study found that the sarkosyl-insoluble proteomes of Parkinson's disease and multiple system atrophy have a significant overlap, consisting mainly of mitochondrial and neuronal synaptic proteins. Other fibrillization-prone proteins possibly cross-seeded by alpha-syn are not found in these extracts.

Oligodendrocyte precursor cells sculpt the visual system by regulating axonal remodeling

Summary: Oligodendrocyte precursor cells have a regulatory role in the precise formation of retinal ganglion cell arbors and visual processing. They can fine-tune neural circuits independently of their canonical role to make myelin.

α-Synuclein Fibrils as Penrose Machines: A Chameleon in the Gear

Francesca De Giorgi et al.

Summary: The article discusses Lionel Penrose's invention of the first self-replicating mechanical device in 1957 and its function, as well as its relevance to the genesis and proliferation of amyloid fibrils. It also highlights the significance of studies on alpha-Synuclein and its similarities to prions, its fibrillization-prone domain, and its nature as an intrinsically disordered protein. Combining these discoveries with the concept of the Penrose machine, the article proposes an explanation for the emergence and spread of different alpha-Synuclein fibril strains in alpha-Synucleinopathies.

Neurons with Cat's Eyes: A Synthetic Strain of α-Synuclein Fibrils Seeding Neuronal Intranuclear Inclusions

Francesca De Giorgi et al.

Summary: The distinct neuropathological features and inclusion bodies in different alpha-Synucleinopathies reflect the strain diversity of invasive alpha-Syn amyloids. However, the contribution of non-amyloid factors to specific diseases is still uncertain. In Multiple System Atrophy, alpha-Syn inclusions in oligodendrocytes seem to result from the storage of amyloid aggregates pre-assembled in neurons. These inclusions originate from unique molecular properties or other factors dysregulated in MSA.

Oligodendrocyte precursor cells ingest axons in the mouse neocortex

JoAnn Buchanan et al.

Summary: Neurons in the developing brain undergo structural refinement through the engulfment and degradation of axonal branches and synapses by glial cells. In the mouse visual cortex, oligodendrocyte precursor cells (OPCs) were found to surround small branches of axons and engage in axon pruning. Compared to microglia, OPCs were more abundant in phagosomes and phagolysosomes, suggesting their substantial contribution to neuronal circuit refinement during cortical development.

Myelin: A gatekeeper of activity-dependent circuit plasticity?

Giulia Bonetto et al.

Summary: The brain's response to changing environments has been primarily studied in terms of functional and structural changes at neurochemical synapses, with less focus on the impact of activity-dependent myelination on neural plasticity and behavior control circuits. Emerging evidence suggests that myelin plasticity plays a significant role in strengthening circuit functions related to learning and behavior.

Neuronal and Glial Distribution of Tau Protein in the Adult Rat and Monkey

Nicholas M. Kanaan et al.

Summary: Tau is a versatile protein found in various cell types in the adult CNS, including neurons and mature oligodendrocytes. Different antibodies used for labeling tau show varied results, indicating the complexity of tau distribution and the need for careful interpretation of experimental data. The findings provide insights into the normal distribution of tau and its role in different compartments within the brain under both physiological and diseased conditions.

Robust α-synuclein pathology in select brainstem neuronal populations is a potential instigator of multiple system atrophy

Ethan W. Hass et al.

Summary: Multiple system atrophy (MSA) is a neurodegenerative disease characterized by the accumulation of glial cytoplasmic inclusions (GCIs). Research has shown that MSA patients have abundant neuronal alpha Syn pathology in specific regions of the brain, which differs from GCIs and may play a role in driving the pathogenesis of MSA.
