Mapping the Complete Photocycle that Powers a Large Stokes Shift Red Fluorescent Protein

Summary: This study utilized femtosecond stimulated Raman spectroscopy and various time-resolved electronic spectroscopies, aided by quantum calculations, to investigate the red fluorescent protein mKeima with a large Stokes shift. It revealed the photocycle and isomerization process of mKeima, and identified the trans-isomer as the dominant contributor to the large Stokes shift emission.

Signal transduction in light-oxygen-voltage receptors lacking the active-site glutamine

Summary: This study investigates whether light-oxygen-voltage (LOV) photoreceptors can transduce light signals in the absence of a conserved glutamine residue. Results show that LOV photoreceptors can still generate effective signaling responses even without this key glutamine. Further structural analysis reveals highly similar light-induced conformational changes regardless of the presence of the glutamine. These findings have implications for biotechnological applications and suggest the evolutionary descent of LOV photoreceptors from redox-active flavoproteins.

Excited State Resonance Raman of Flavin Mononucleotide: Comparison of Theory and Experiment

Summary: Flavoproteins that absorb blue light play crucial roles in various photobiological processes. Detailed studies on the excited state structure and dynamics have been conducted, especially through time-resolved vibrational spectroscopy. Although there is generally good agreement between calculated and observed enhancements, some prominently enhanced bands are absent from the calculations, indicating the need for further theoretical development.

Summary: This study computationally investigated three distinct LOV domains, uncovering some mechanistic insights of the LOV photocycle by constructing extended protein-chromophore models and calculating rotational barriers and energies of critical residues. The agreement between experimental and computational methods allowed for the assignment of observed bands and the identification of a facile glutamine flip at the adduct intermediate, providing new clues about early events in signal transduction of LOV proteins.
