Differential analyte derivatization enables unbiased MALDI-TOF-based high-throughput screening: A proof-of-concept study for the discovery of catechol-o-methyltransferase inhibitors

Martin Winter et al.

Summary: Recent advances in label-free high-throughput screening using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) have provided unprecedented opportunities for drug discovery. This technology allows for direct quantification of analytes with high precision and robustness. Researchers have developed a method to detect 3-methoxytyramine, a physiological enzyme product, by expanding the range of analytes that can be addressed via MALDI-TOF HTS. They have also devised a differential derivatization workflow to address the issue of isobaric interference in MALDI-TOF assays. Furthermore, they report on the first label-free HTS campaign for the identification of COMT inhibitors using MALDI-TOF-MS analysis in a miniaturized format.

Cytotoxic CD8+T cells in cancer and cancer immunotherapy

Hans Raskov et al.

Summary: The functions and interactions between the innate and adaptive immune systems play a crucial role in anticancer immunity. Cytotoxic T cells with CD8 as the most effective effectors in the anticancer immune response form the basis of successful cancer immunotherapies. Advancements in genetically modified or synthetic receptors on cytotoxic T cells are being developed for future cancer treatment. Combinatory regimens may optimize treatment effects and reduce adverse events in cancer therapy.

High-Throughput Mass Spectrometry for Hit Identification: Current Landscape and Future Perspectives

David G. McLaren et al.

Summary: Mass spectrometry has been widely used as a label-free, direct-detection method for various therapeutically relevant target classes for nearly two decades. Different mass spectrometry platforms offer unique strengths and performance characteristics when it comes to high-throughput screening, with sensitivity, specificity, wide dynamic range, and adaptability to complex matrices being key advantages. By leveraging these inherent strengths, mass spectrometry can enhance the discriminating power and physiological relevance of assays in screening cascades for the search of hits, leads, and drugs.

Common allotypes of ER aminopeptidase 1 have substrate-dependent and highly variable enzymatic properties

Jonathan P. Hutchinson et al.

Summary: The study revealed that ERAP1 allotypes exhibit a wide range of enzymatic activities, with one allotype, associated with Behcet's disease, consistently showing low activity outlier. Through in vitro analysis, it was found that ERAP1 allotypes can differ in both catalytic efficiency and substrate affinity, leading to changes in intermediate accumulation in multistep trimming reactions.
