Oridonin interrupts cellular bioenergetics to suppress glioma cell growth by down-regulating PCK2

Summary: Oridonin was found to significantly inhibit the growth of glioma cells in vitro and in vivo by reducing cellular metabolism. The down-regulation of PCK2 was identified as a potential mechanism of Oridonin's anti-tumor properties in glioma, as confirmed by the effects of PCK2 depletion mimicking those of Oridonin treatment. Additionally, PCK2 up-regulation in higher grade glioma was associated with poor outcomes, suggesting its potential as a therapeutic target.
